Abstract
Teški metali su postojani zagađivači okoliša i mogu imati toksičan učinak. Zbog sklonosti akumulaciji opasni su za žive organizme. Djelovanje pojedinih teških metala na različitim razinama biološke organizacije uglavnom je dobro poznato, dok je kombinirano djelovanje više teških metala na organizam još nedovoljno istraženo. U ovom je radu istražen genotoksični učinak kadmija i bakra, pojedinačno i u kombinacijama, u klijancima i odraslim biljkama duhana Nicotiana tabacum L. Cilj rada bio je odrediti polimorfizam molekule DNA kao posljedicu izlaganja duhana teškim metalima kadmiju i bakru, te utvrditi djeluju li ti metali međusobno sinergistički ili antagonistički. Analiza sadržaja metala pokazala je da učinak bakra i kadmija ovisi o koncentraciji metala u biljci, ali i o razvojnom stadiju biljke. Bakar je u klijancima povećao toksične učinke kadmija (sinergistički učinak), dok je kod odraslih biljaka smanjio toksičan učinak kadmija (antagonistički učinak). Klijanci su u odnosu na odrasle biljke imali smanjenu stabilnost genoma.
Abstract (english)
Heavy metals are persistent environmental pollutants, and can have toxic effects due to their accumulation in living organisms. Effect of heavy metals at different levels of biological organization is generally well known, but the combined effect of several heavy metals on the organism has been insufficiently studied. The genotoxic effect of cadmium and copper, alone and in combination, in tobacco Nicotiana tabacum L., was investigate in this study. The aim was to determine the polymorphism of DNA as a result of tobacco exposure to heavy metals cadmium and copper, and evaluate if the combined effect of these metals is synergistic or antagonistic. Metal content analysis revealed that the influence of copper and cadmium depended on the concentration and developmental stage of plants. Copper increased toxic effects of cadmium in the seedlings (synergistic effect), whereas in adult plants reduce toxic effects of cadmium (antagonistic effect). Seedlings have reduced genome stability in comparison to adult plants.
