Abstract
Citomegalovirusi (CMV) spadaju u skupinu betaherpesvirusa te posjeduju strogu specifičnost u smislu vrste koju mogu inficirati. To onemogućava istraživanje učinka humane CMV (HCMV) infekcije na životinjama. HCMV i mišji CMV (MCMV) u svom genomu posjeduju više gena čija je svrha negativna regulacija imunološkog odgovora domaćina. Primijećeno je da dolazi i do smanjenja ekspresije liganda za imunološke receptore, molekule PVR (poliovirusni receptor, CD155), na površini stanica inficiranih HCMV-om. Cilj ovog istraživanja bio je odrediti gene MCMV-a odgovorne za imunoregulaciju, proizvesti virusne proteine potencijalne regulatore molekule PVR i uspostaviti metode za izučavanje interakcije PVR/TIGIT. Rezultati dobiveni biokemijskom analizom pokazali su regulacijsko djelovanje proteina kodiranih regijom m19-m21 na molekulu PVR u stanicama mišjih fibroblasta inficiranih MCMV-om. U ekspresijskom sustavu E. coli uspješno je proizveden protein m21. Protein TIGIT koji sudjeluje u regulaciji imunološkog odgovora vezanjem na PVR uspješno je izoliran iz transficiranih staničnih linija.
Abstract (english)
Cytomegalovirus (CMV) is a betaherpesvirus and thus strictly species specific. This prevents the research of human CMV (HCMV) infection in animals. HCMV and murine CMV (MCMV) possess several genes in their genome whose purpose is to negatively regulate immune responses of the host. In line with this, the reduction of surface expression of PVR (poliovirus receptor, CD155), a ligand for immune receptors, was observed on the surface of HCMV infected cells. The aim of this study was to determine MCMV genes that are responsible for the PVR regulation, to produce viral proteins which regulate the expression of PVR molecules and to establish methods for studying the PVR/TIGIT interaction. Biochemical analysis has shown the capacity of viral proteins encoded by m19-m21 region to regulate PVR molecules in MCMV infected mouse fibroblasts. The protein m21 was successfully produced in E. coli expression system. Immune receptor TIGIT that participates in the regulation of immune responses by binding to PVR was successfully isolated from transfected cell lines.
