Abstract
In vitro testiranja pojedinih pesticida najbrži su i najjednostavniji način za utvrđivanje toksičnog potencijala pojedinih pesticida. Cilj rada je utvrditi na koji način pesticidi imazalil, cipermetrin i karbendazim te njihove kombinacije utječu na hemolizu na eritrocitnom modelu in vitro u usporedbi sa hematotoksičnim potencijalom in vivo u Swiss-miša. Hemolitički potencijal u in vitro pokusu utvrdio se modificiranom metodom po Grinbergu. Eritrociti dobiveni iz repne vene miša isprani su te resuspendirani (2% v/v) u izotoničnoj otopini PBS-a s 5.5 mM glukozom. U svaku otopinu dodane su otopine pesticida i njihovih kombinacija od 5 μM do 10 mM. Postotak hemolize u vremenu određen je spektrofotometrijski. U in vivo pokusu životinje su izložene NOAEL dozama pojedinog pesticida i kombinacijama (NOAEL, 1:1). Pesticidi su svakodnevno aplicirani oralno tijekom 28 dana. Utvrđeni su hematološki parametri: brojnost eritrocita RBC, hematokrit HCT, hemoglobin HB. Imazalil i njegove kombinacije pokazuju najjači hemolitički potencijal u in vitro pokusu u vremenu u svim koncentracijama. U in vivo pokusu, nakon 28 dana tretman imazalilom i cipermetrinom rezultirao je policitemijom. Karbendazim nije značajno promijenio broj eritrocita.
Abstract (english)
In vitro assays of pesticides are the fastest and easiest way to determine the toxic potential of certain pesticides. The aim was to determine how the pesticides imazalil, cypermethrine and carbendazime and their combinations influence erythrocyte hemolysis in vitro compared with hematotoxic potential in vivo in Swiss-mouse. Hemolytic potential in a in vitro experiment was measured with the modified method according to Grinberg. Erythrocytes obtained from mouse tail veins were washed and resuspended (2% v / v) in isotonic solution of PBS with 5.5 mm glucose. In each solution pesticides and their combination were added from 5 μM to 10 mM. The percentage of hemolysis was determined spectrophotometrically. In in vivo experiment animals were exposed to the NOAEL doses of the each individual pesticide and their combinations (NOAEL, 1:1). Pesticides were applied orally during 28 consecutive days. Following hematological parameters were determined: the red blood cell count RBC, HCT hematocrit, hemoglobin HB. Imazalil and its combinations showed the strongest hemolytic potential in in vitro experiment at all concentrations. In in vivo experiment, after 28 days treatment with imazalil and cypermethrine caused polycythemia. Carbendazime did not significantly change the number of red blood cells.
