Abstract
Transkripcijom se gen sadržan u molekuli DNA prepisuje pomoću enzima RNA-polimeraze II u molekulu glasničke RNA, te zatim prevodi u proteine. Transkripcija određenih gena povezana je s hranjivim tvarima u okolini. Aktivator transkripcije Gal4 regulira gene koji sudjeluju u metabolizmu galaktoze. U ovom radu koristi se i skraćeni Gal4, sastavljen od domene za vezanje DNA i minimalne aktivacijske domene. Gal4 različitim afinitetom veže promotorske regije gena u kojima prepoznaje specifičan slijed DNA. Protein-kinaza Srb10 jedna je od podjedinica kompleksa Medijator, te regulira transkripciju. Ovim radom određene su razlike u razini transkripcije u sojevima NLY2 SV15, NLY2 SV15Δsrb10, NLY2 Mel1p, NLY2 Mel1pΔsrb10, NLY2 RY171 i NLY2 RY171Δsrb10 u neinducibilnim i inducibilnim uvjetima, koristeći nativni i kratki oblik aktivatora Gal4. Razina transkripcije gena pokazatelja u ovim sojevima određena je testom β-galaktozidaze, a endogenog gena mel1 testom α-galaktozidaze. U ovom radu je pokazano da protein-kinaza Srb10 može regulirati transkripciju neovisno o S699. Protein-kinaza Srb10(CDK8) regulira transkripciju koordinacijom signala u skladu sa uvjetima u okolišu preko interakcija sa transkripcijskom mašinerijom, koaktivatorima, te regulacijom aktivacije i obrtaja transkripcijskog aktivatora Gal4.
Abstract (english)
Transcription is a process of transcibing a gene within a DNA molecule into a messenger RNA molecule using RNA-polymerase II. The level of transcription of certain genes is related to available nutrients. The Gal4 transcriptional activator is responsible for transcriptional regulation of genes involved galactose metabolism. A short form of Gal4, consisting of only the DNA binding and minimal activation domains is used in this study. Gal4 binds to specific DNA sequences within various gene promotor regions with different binding affinity. The Protein-kinase Srb10 (CDK8) is one of the subunits of Mediator complex, and it is involved in transcription regulation. This thesis determined the differeces in the level of transcription using NLY2 SV15, NLY2 SV15Δsrb10, NLY2 Mel1p, NLY2 Mel1pΔsrb10, NLY2 RY171 and NLY2 RY171Δsrb10 yeast trains, in inducible and repressive conditions, using the short and the native form of the transcription activator Gal4, attempting to determine the role of the protein-kinase Srb10(CDK8) based on obtained data. The level of transcription of reporter gene is determined by β-galactosidase assay, and of endogenous mel1 gene by α-galactosidase assay. In this thesis it is shown that protein-kinase Srb10 regulaes transcription independently of the S699. Protein-kinase Srb(CDK8) regulates transcription by coordinating signals according to environmental conditions, interacting with the transcription machinery, transcriptional co-activators and by regulating Gal4 activation and turnover.
