Abstract
Retrotranspozon LINE-1 jedini je aktivan pokretni genetički element kod čovjeka. Pokretanje elementa LINE-1 može dovesti do genomske nestabilnosti i nastanka bolesti. U stanicama postoje brojni mehanizmi kojima se njegovo pokretanje nastoji spriječiti, pri čemu je najvažnije epigenetičko utišavanje transkripcije elementa LINE-1. Njegova ekspresija česta je u stanicama tumora i može dovesti do progresije bolesti. Mutacije koje uzrokuje pokretanje elementa LINE1 u tumorskim stanicama su insercije, delecije i kromosomski rearanžmani. Također, ekspresija LINE-1 može poremetiti regulaciju ekspresije drugih gena. Postoji zanimljiva poveznica između ekspresije pokretnih genetičkih elemenata i nastanka autoimunih bolesti. Stanični senzori nukleinskih kiselina mogu prepoznati element LINE-1 na isti način kao što prepoznaju virusne nukleinske kiseline i pokrenuti imunosni odgovor.
Abstract (english)
The LINE-1 retrotransposon is the only active mobile genetic element in humans. Activation of the LINE-1 element can lead to genomic instability and the development of diseases. Cells employ various mechanisms to prevent its activation, with the most important being epigenetic regulation of transcription. The expression of the LINE-1 element is common in tumor cells and can contribute to progression of disease. Mutations caused by the activation of the LINE-1 element in tumor cells include insertions, deletions, and chromosomal rearrangements. Additionally, LINE-1 expression can disrupt the regulation of other genes. Moreover, there is a link between expression of transposable elements and the development of autoimmune diseases. Cellular sensors of nucleic acids can recognize the LINE-1 element in the same way they recognize viral nucleic acids, triggering an immune response.
