Abstract
Multipla skleroza je autoimuna i neurodegenerativna bolest središnjeg živčanog sustava. Zbog neregulirane funkcije imunosnog sustava, dolazi do demijelinizacije aksona koju prate upalni procesi i naposljetku propadanje neurona. Uzročnik bolesti još nije izdvojen, dok je za početak upale potrebna genska predispozicija kao i vanjski okolišni čimbenik koji služi kao okidač. Vanjskih čimbenika je puno, a uključuju virusne infekcije, nedostatak vitamina D, prekomjernu tjelesnu težinu, pušenje, ali i promjene u mikrobioti i epigenetičke modifikacije. Liječenje, u većini slučajeva, uključuje imunomodulaciju kojom se izaziva niska razina limfocita. Međutim, i dalje ne postoji lijek koji sprječava neurodegeneraciju pa se liječenje i dalje bazira samo na ublažavanju simptoma.
Abstract (english)
Multiple sclerosis is an autoimmune and neurodegenerative disease that affects the central nervous system. Dysregulated immune system causes demyelination of axons, which is accompanied with inflammation and, finally, death of neurons. The cause of the disease still hasn’t been singled out, even though, both genetic predisposition and environmental risk factors are needed for the onset. There are many environmental risk factors, including viral infections, vitamin D deficiency, overweight, smoking, and also the changes in gut microbiota and epigenetic modifications. Treatment is, in most cases, immunomodulation that causes low lymphocyte levels. However, a cure that would prevent neurodegeneration still hasn’t been found, so the treatment is based solely on symptoms relief.
