Abstract
N glikozilacij a u šećernoj bolesti tipa 1 (ŠBT1) povezan a je s početkom bolesti , dijabetičkim komplikacijama i lošom glikemijskom kontrolom , a osim navedenog, novija istraživanja naglašavaju ulogu i munoglobulin a A (IgA) i njegove glikozilacije u autoimunim bolestima. U ovom istraživanju je tekućinsk om kromatografij om ultra visoke djelotv ornosti analizirana povezanost N glikoma ukupnih serumskih proteina te antitijela IgA s a ŠBT1 kako bi se pružio uvid u obrasce N glikozilacije odraslih, identificirale promjene jedinstvene za početak bolesti u djece , te ispitala povezanost N glikoma s komplikacijama i rizičnim faktorima N glikom ukupnih serumskih proteina značajno se mijenja u ŠBT1 u obrazac nalik upalnim procesima te pokazuje asocijacije s glikemijskom kontrolom i navikama pušenja, ali ne sa statusom komplikacija Početak ŠBT1 u djece slabo se odražava na N glikom antitijela IgA, dok je ŠBT1 u odraslih povezan a s brojnim promjenama i povećanim grananjem N glika n a . Navedeno upućuj e na uključenost N glikoma u različitim fazama ŠBT1 te na potencijalnu ulogu u imunosnim odgovorima i upalnim procesima u ŠBT1
Abstract (english)
N-glycosylation in type 1 diabetes (T1D) is linked to disease onset, complications, and poor glycaemic control. In addition, recent findings emphasize the role of immunoglobulin A (IgA) and its glycosylation in autoimmune diseases. In this study, the association of N-glycome of total serum proteins and IgA with T1D was analysed by ultra-high performance liquid chromatography to elucidate the N-glycosylation patterns of adults, identify changes unique to the onset in children, and examine the association of N-glycome with complications and risk factors. N-glycome of total serum proteins significantly changes in T1D in an inflammatory-like pattern, and associates with glycaemic control and smoking, but not complications. The onset of T1D barely reflects in the N-glycome of IgA, while T1D in adults associates with substantial changes and increased branching. These findings indicate the involvement of N-glycome in different stages of T1D and a potential role in immune responses and inflammatory processes in T1D.
