Abstract
Adenokarcinom pluća jedan je od najčešćih malignih tumora današnjice. Za njega je specifična mutacija gena EGFR na temelju koje se dijeli u podskupine za koje su razvijene ciljane terapije. Kao i svi tumori, adenokarcinom pluća uzrokuje imunosni odgovor domaćina promjenama vlastitog transkriptoma, ali i promjenama koje su uzrokovane infekcijom tumorskih stanica virusima, poput one citomegalovirusom (HCMV). Cilj ovog istraživanja bio je utvrditi potencijalne razlike u imunosnom odgovoru kod podskupina adenokarcinoma pluća sa i bez mutacija u genu EGFR, potaknute promjenama transkriptoma tumorskih stanica ili prisutnošću HCMV-a u njima. U skupini adenokarcinoma pluća bez mutacija u genu EGFR utvrđena je značajno povišena ekspresija gena IL-1β i CXCR4 u odnosu na skupinu adenokarcinoma pluća s mutacijama u genu EGFR. Utvrđena je i značajna povezanost između povišene ekspresije gena CXCR4 i infekcije tumorskih stanica HCMV-om kod adenokarcinoma pluća bez mutacija u genu EGFR. Razlike u imunosnom odgovoru između ispitivanih skupina adenokarcinoma pluća nisu nađene.
Abstract (english)
Lung adenocarcinoma is one of the most common malignant tumors today. The EGFR gene mutations are specific for lung adenocarcinoma, therefore it is divided into subgroups for which targeted therapies have been developed. Like all tumors, lung adenocarcinoma stimulates the host's immune response to changes in its transcriptome and changes caused by viral infection of tumor cells such as cytomegalovirus (HCMV). This study aimed to determine potential differences in the immune response in subgroups of lung adenocarcinomas with and without EGFR gene mutations caused by changes in the transcriptome of tumor cells or the presence of HCMV. In the group of lung adenocarcinoma without EGFR gene mutations, a significantly elevated expression of IL-1β and CXCR4 genes was found compared to the group of lung adenocarcinoma with EGFR gene mutations. A significant association was also determined between elevated CXCR4 gene expression and HCMV infection of tumor cells in lung adenocarcinoma without EGFR gene mutations. No differences were found in the immune response between the analyzed groups of lung adenocarcinoma.
