Abstract
Zbog različitih molekulskih konformacija i međumolekulskih interakcija, čvrsti oblici iste djelatne tvari mogu pokazivati različita fizikalna i biofarmaceutska svojstva. Kako bi se poboljšala stabilnost i učinkovitost lijeka, djelatna se tvar često prevodi u čvrstu formu soli. U okviru ovog rada, pripravljene su izučene u čvrstom stanju odabrane soli dvaju izomera piridinkarbohidrazida, od kojih je jedan – piridin-4-karbohidrazid ili izoniazid – poznati antibiotik koji se primjenjuje u obliku slobodne baze. Ispitani su uvjeti priprave i kristalizacije klorida, bromida, nitrata, perklorata, hidrogensulfata, sulfata i dihidrogenfosfata navedenih karbohidrazida. Pripravljenim solima određene su strukture u čvrstom stanju na temelju podataka difrakcije rentgenskog zračenja na jediničnom kristalu i na praškastom uzorku. Pripravljene soli okarakterizirane su i termičim metodama analize, kao i FT-IR spektroskopijom. Konačno, utvrđen je empirijski suodnos između strukture i svojstava pripravljenih soli.
Abstract (english)
Due to different molecular conformations and intermolecular interactions, solid forms of the same active substance can exhibit different physical and biopharmaceutical properties. To improve the stability and efficacy of a drug, the active substance is often converted into a solid salt form. In this thesis, the selected salts of two isomers of pyridinecarboxhydrazide, one of which is — pyridine-4-carbohydrazide or isoniazid — a known antibiotic administered in the form of a free base, were prepared and studied in the solid-state. The conditions for preparing and crystallizing the chlorides, bromides, nitrates, perchlorates, hydrogen sulfates, sulfates, and dihydrogen phosphates of these carbohydrazides were studied and defined. Solid-state structures of the prepared salts were determined based on X-ray diffraction data on the single- crystal and powder samples. The prepared salts were also characterized by thermal analysis as well as FT-IR spectroscopy. Finally, an empirical correlation between the structure and properties of the prepared salts was established.
