Abstract
Neuropeptid Y (NPY) izražavaju stanice iz loze osteoblasta. In vitro studije su pokazale da NPY ima negativan učinak na diferencijaciju osteoblasta. Vođeni pretpostavkom da NPY izražen u stanicama loze zrelih osteoblasta može kontrolirati diferencijaciju mezenhimskih progenitorskih stanica istražili smo in vivo ulogu neuropeptida Y kao regulatora osteogeneze. U tu svrhu uzgojili smo transgeničnog miša (Col2.3NPY-ires GFPtopaz). Lančana reakcija polimerazom u stvarnom vremenu (RT-qPCR) i NPY EIA analiza pokazali su pojačani izražaj NPY-a u transgeničnim miševima. Col2.3NPY miševi imali su 25% smanjen udio trabekularne kosti i 7-10% tanje trabekule. Analiza genske ekspresije nije pokazala nikakve razlike u ekspresiji OC, BSP i DMP-1 kod transgeničnih životinja. Col2.3NPY mCOB kulture imale su 6 puta manju površinu mineralizacije te smanjeni izražaj markera osteogeneze. Ovaj mišji model pokazao je da lokalno izražen NPY ima važnu ulogu u regulaciji koštane mase te regulira promjene u koštanoj masi. Svi ovi rezultati pokazali su da periferni signalni put NPY-a ima bitnu ulogu u koštanoj pregradnji.
Abstract (english)
Neuropeptide Y is expressed by osteoblast lineage cells and when administered in vitro can inhibit osteoblast lineage differentiation. We hypothesize that NPY, generated by mature osteoblast lineage cells, can control and modulate differentiation of osteoprogenitor cells. In this study we have evaluated the effects of bone-specific overexpression by generating Col2.3NPY-ires GFPtopaz transgenic mice. RT-qPCR and NPY EIA analysis showed increased expression of NPY in transgenic mice. Col2.3NPY mice had 25% lower trabecular volume and 7-10% lower trabecular thickness. Gene expression analysis in Col2.3NPY mice did not show any changes in expression levels of OC, BSP and DMP-1. Terminal osteoblast differentiation was impaired in Col2.3NPY mCOB cultures as indicated by a 6-fold decrease in mineralized area and decreased expression of all osteogenic markers. Our model has shown that locally expressed NPY plays an important role in regulation of bone mass. All these findings suggest that peripheral mechanism of the NPY signaling is important in bone remodeling.
