| Abstract | Oscilatomo gibanje jezgre stanice S. pombe pred mejozu nužno je za pravilno kromosomsko sparivanje, rekombinaciju genetskog materijala i održivost spora. Vukući mikrotubule dineinski motor uzrokuje navedene oscilacije, gibajući pri tome jezgru naprijed-nazad unutar stanice. Dosadašnji teorijski model aproksimacije srednjeg polja (Vogel et al., 2009) opisuje jezgrine oscilacije bazirajući se na otpuštanju motora s mikrotubula pod utjecajem sile opterećenja te njihovom vezivanju na mikrotubule ovisnom o duljini mikrotubula. Model pretpostavlja velik ukupan broj motora unutar stanice te se služi aproksimacijom srednjeg polja. No, same fluktuacije broja motora su zanemarene. Za mali ukupni broj motora u stanici, ove fluktuacije su značajne te dovode do nepravilnih jezgrinih oscilacija. Kako bismo provjerili događaju li se pravilne oscilacije jezgre uz fluktuaciju broja motora izveli smo računalne stohastičke simulacije temeljene na promatranju ponašanja pojedinog dineinskog motora. Dinamiku sustava opisali smo „master" jednadžbom određujući pri tome vjerojatnost P(N1, Nn t) pronalaženja N1, odnosno Nr motora vezanih na lijevi, odnosno desni mikrotubul u trenutku t. Budući da postavljenu „master" jednadžbu nije moguće riješiti analitički, rezultate smo ostvarili numeričkim rješavanjem koristeći pri tome Gillespie algoritam. Izveli smo stohastičke simulacije za različit ukupan broj motora u citoplazmatskom rezervoaru, počevši od velikih vrijednosti broja motora pa sve do samo jednog motora u rezervoaru. Pri velikom broju motora stohastički model pruža dobru podudarnost s modelom aproksimacije srednjeg polja. Nasuprot tome, smanjivanjem ukupnog broja motora, oscilacije gube na periodičnosti te aproksimacija srednjeg polja više nije valjana. Metodom autokorelacije utvrdili smo optimalan raspon broja motora u rezervoaru potreban za pravilne periodične oscilacije jezgre, Nuk = 20 - 40, koji se slaže s eksperimentalnim mjerenjima. Ustvrdili smo ovisnost raspona optimalnog broja motora o trenju sustava te smo na kraju ispitali parametarski prostor stope priključivanja kon te konstante otpuštanja koff. S obzirom da je citoplazmatski dinein molekularni motor prisutan u mnogim eukariotskim stanicama, okvir koji se koristi u ovom radu ima veliku mogućnost primjene i u drugim organizmima, uključujući i ljude. |
| Abstract (english) | During the meiotic prophase of the life cycle of fission yeast, oscillatory nuclear movement is crucial for proper chromosome pairing, recombination and spore viability. This movement is driven by dynein motors pulling on microtubules, consequently moving the nucleus back and forth inside the cell. A theoretical model (V ogel et a!„ 2009) of nuclear oscillations, based on load-dependent detachrnent and microtubule length-dependent attachment of the motors, describes nuclear oscillations using large number of motors and a mean-field approximation. However, fluctuations in the number of active force generators are neglected. For small number of motors these fluctuations become important and lead to noisy oscillations. To verify that oscillations still occur in the presence of force fluctuations, we have performed stochastic computer simulations of individual force generators. We bave represented the dynamics of the system by a master equation for the probability P(N1 ,NT ,t) of finding a configuration of N1 and NT particles attached, both to the cell cortex and to the microtubule on the left and the right side of SPB, respectively, at time t. We have obtained our results by solving the equation numerically using Gillespie's algorithm. We have performed computer simulations for a different number of motors, starting with a large number of motors and ending with the situation where only one motor is present. In the limit of a large number of motors, we obtain results similar to those suggested by the mean-field approximation. On the contrary, as the number of particles decreases, the mean-field approximation is no longer valid. Using the method of autocorrelation we have determined the number of motors necessary for robust nucleus oscillations, Nuk = 20 - 40. We investigated how does friction of system influence oscillatory movement of nucleus. Last, we checked parametric space for attachrnent rate kon and constant of detachment ko. Comparing the results obtained by stochastic simulations to those measured experimentally, we have confirmed validity of our model which can now be used in other scientific experiments conducted on similar biological systems. As the cytoplasmic dynein is found in many eukaryotic cells, the frame that is used in this paper has a number of applications in other organisms, including humans. |
