Abstract
Magnetske nanočestice željezovih oksida (magnetske NČ) spadaju u važnu skupinu materijala s primjenom u katalizi i biomedicini. Magnetske NČ se koriste kao kontrastna sredstva u oslikavanju magnetskom rezonancijom, kao nosioci lijekova za njihovo kontrolirano otpuštanje i kao terapijska sredstva za uništavanje stanica raka postupkom hipertermije. δ-FeOOH je slabo kristalni materijal koji ima veliku specifičnu površinu, veliki broj hidroksilnih skupina i mnogo bolja adsorpcijska svojstva u odnosu na nanočestice magnetita i maghemita. Magnetske NČ δ-FeOOH nedavno su sintetizirane radiolitičkim postupkom, međutim citoksičnost tako sintetiziranih NČ δ-FeOOH do sada nije ispitivana. U ovom diplomskom radu magnetske NČ δ-FeOOH sintetizirane su γ-ozračivanjem (radiolitičkom sintezom). Sintetizirane NČ δ-FeOOH aplicirane su na embrionalne stanice bubrega čovjeka HEK 293, a vijabilnost stanica ispitana je testom MTT. Test MTT rađen je 24 sata i 48 sati nakon tretiranja HEK 293 stanica nanočesticama δ-FeOOH. Rezultati dobiveni invertnim mikroskopom pokazali su da nanočestice δ-FeOOH ulaze u stanice i da agregiraju unutar njih. Također, rezultati dobiveni invertnim mikroskopom u skladu su s rezultatima testa MTT, odnosno u oba slučaja HEK 293 stanice su pokazale dobru vijabilnost nakon izlaganja nanočesticama δ-FeOOH u koncentracijskom rasponu od 0,95 x 1011 do 1,9 x 1012 NČ L-1. Novi radiolitički način sinteze magnetskih NČ δ-FeOOH i potvrda njihove netoksičnosti snažan je poticaj za istraživanja njihove moguće primjene u biomedicini.
Abstract (english)
Magnetic iron oxide nanoparticles (magnetic NPs) belong to an important group of materials with applications in catalysis and biomedicine. Magnetic NPs are used as contrast media in magnetic resonance imaging, as carriers of controlled release drugs, and as therapeutic agents for the eradication of cancer cells by hyperthermia. δ-FeOOH is a weak crystalline material having a large specific surface, a large number of hydroxyl groups, and much better adsorption properties than magnetite and maghemite NPs. Recently, δ-FeOOH NPs have been radiolytically synthesized, however, the cytotoxicity of thus synthesized δ-FeOOH NPs has not been investigated so far. In this graduate thesis the magnetic δ-FeOOH NPs were synthesized using γ-irradiation (radiolytical synthesis). Synthesised δ-FeOOH NPs were applied to human embryonic kidney cells HEK 293, and cell viability was tested for MTT. Cell monitoring was tested by MTT test for 24 hours and 48 hours after treatment with δ-FeOOH nanoparticles. The results obtained by an invert microscope showed that δ-FeOOH NPs entered and aggregated within the cells. Besides, the results obtained by the invert microscope are in accordance with the MTT test results, i.e. in both cases HEK 293 cells showed quite good viability after exposure to δ-FeOOH NPS at a concentration range of 0.95 x 1011 to 1.9 x 1012 NPs L-1. The new radiolytical synthesis of δ-FeOOH NPs and their non-toxicity verification have shown that δ-FeOOH NPs possess great potential for use in biomedical applications.
