Abstract
U okviru ovog diplomskog rada opisana je kemijska sinteza amfifilnog derivata ßciklodekstrina, heptakis[6-deoksi-6-dodeciltio-2-O-oligo-(etilen glikol)]-ß-ciklodekstrina (EO-CD-SR) i priprava funkcionalnih supramolekulskih sustava s adamantanskim gvanidinima. Priređeni su i karakterizirani supramolekulski sustavi adamantanskih gvanidina s liposomima građenim od fosfatidilkolina, liposomima u koje je ugrađen amfifilni derivat ciklodekstrina te ciklodekstrinskim vezikulama građenim samo od amfifilnog derivata ciklodekstrina. Efikasnost ugrađivanja adamantanskih gvanidina u priređene supramolekulske sustave određena je spektrofotometrijski, a veličina i površinski naboj određeni su metodom dinamičkog raspršenja svjetlosti. Nadalje, opisano je nastajanje kompleksa između adamantanskih gvanidina i ß-ciklodekstrina metodom spektralne izmjene te je izračunata konstanta vezanja Ka' za sve testirane adamantanske gvanidine AG 1-5. Interakcije između priređenih kompleksa liposoma i ciklodekstrinskih vezikula s adamantanskim gvanidinima i fluorescentno obiljeţene DNA potvrđene su metodom fluorescencijske korelacijske spektroskopije.
Abstract (english)
In this graduate thesis the chemical synthesis of β-cyclodextrin, heptakis [6-deoxy-6- dodecylthio-2-O-oligo-(ethylene glycol)]-ß-cyclodextrin (EO-CD-SR) and preparation of functional supramolecular systems with adamantanes guanidine are described. Supramolecular systems of adamantanes guanidine with phosphatidylcholine liposomes, liposomes incorporating EO-CD-SR derivative, and cyclodextrin vesicles composed only from the amphiphilic cyclodextrin derivative were prepared and characterized. Efficiency of incorporation of adamantanes guanidine into prepared supramolecular systems was measured spectrophotometrically, and the size and surface charge were determined by dynamic light scattering method. Furthermore, the formation of complexes between adamantanes guanidine and ß-cyclodextrin was studied by spectral replacement method while the Ka' binding constant was calculated for all tested AG 1-5 adamantanes guanidine. Interactions of the liposome and cyclodextrin vesicles with adamantanes guanidine complex and fluorescently labelled DNA were confirmed by fluorescence correlation spectroscopy.
