Abstract
Triptofan-sintaza je bifunkcionalni multienzimski kompleks koji katalizira posljednja dva koraka u sintezi triptofana. Triptofan spada u aromatske kiseline te stoga početak sinteze dijeli sa aminokiselinama fenilalaninom i tirozinom. Sintetiziraju ga prokarioti, gljive i biljke. Cilj ovog seminarskog rada je opisati građu triptofan-sintaze, potanko objasniti reakcijski mehanizam na svakoj od podjedinica te pojasniti prijenos informacija između njih. Također, triptofan-sintaza je enzim preko kojeg se mogu shvatiti određena svojstva koja posjeduju i mnogi ostali enzimi a važna su za njihovo efikasno funkcioniranje kao što su alosterička regulacija, prijenos supstrata kroz intramolekulski tunel i biokemija koenzima PLP-a (piridoksal-fosfat). Saznanja o načinu funkcioniranja ove nanomašinerije imaju i direktnu korist u primjerice dizajniranju herbicida ili lijekova protiv različitih bolesti. Usprkos tome što je struktura triptofan-sintaze poznata već dosta dugo vremena, relativno nedavno uporabom sofisticiranijih metoda istraživanja je otkrivena njezina detaljnija struktura. No, još uvijek određene intermedijalne strukture nisu riješene te su stoga potrebna i daljnja istraživanja.
Abstract (english)
Tryptophan-synthase is a multienzyme complex, catalyzing the last two steps in the synthesis of tryptophan. Tryptophan is one of the aromatic acids so it shares the beginning of the synthesis with amino acids phenylalanine and tyrosine. It is synthesized by prokaryotes, fungi and plants. Here we describe the structure of tryptophan-synthase, explain in detail mechanism of the reaction at each subunit and explain the transfer of information between them. Moreover, tryptophan-synthase may be used as a model enzyme to understand certain properties that are present in many other enzymes and are important for their efficiency, like allosteric regulation, substrate channeling through the intramolecular tunnel and biochemistry of coenzyme PLP (pyridoxal-phosphate). The understanding of molecular mechanism of this nano-machine, have direct benefits, for example in design of herbicides or drugs against various diseases. Despite the fact that the structure of tryptophan-synthase is known for a quite long time, relatively recently, because of use of more sophisticated methods of research, details of the structure have been revealed. But, there are still certain intermediate structures that are unknown and therefore, further research is needed.
