Abstract
Ključni molekularni i stanični mehanizmi koji dovode do pojave i progresije karcinoma prostate i jetara još uvijek su u velikoj mjeri nepoznati. U ovom smo radu pokazali da pri uzastopnom precjepljivanju normalnih stanica epitela prostate štakora (PEC) dolazi do spontane transformacije uslijed koje su PEC visokog pasaža (p>85) stekle sposobnost rasta neovisnog o čvrstoj podlozi i stvaranja tumora nakon injekcije u imunodeficijentne miševe. Mala subpopulacija tih stanica (SAI) imala je sposobnost prolaska kroz meki agar te je pokazivala značajne razlike u morfologiji, proliferaciji, pokretljivosti i ekspresiji signalnih proteina kao i veći tumorogeni potencijal. Ispitali smo također razlike između ekstracelularnih vezikula (EV) koje su otpuštale WB-F334 (WB) stanice epitela jetara sa svojstvima matičnih, GP7TB stanice dobivene kemijskom transformacijom WB, te GP7TB.SAI stanice invazivne na mekom agaru. Osim razlika u broju, veličini i proteomičkom sastavu EV pojedinih linija, uočeno je da su EV GP7TB.SAI stimulirale citotoksičnost stanica prirođenih ubojica (NK) nasuprot onima iz GP7TB unatoč tomu što su ove EV pokazale veću ekspresiju površinskog glikoproteina Necl5.
Abstract (english)
The critical molecular and cellular mechanisms involved in the development and progression of prostate and liver cancer remain elusive. In this work we demonstrated that normal rat prostate epithelial cells (PEC) undergo spontaneous transformation at high passage (p>85) evidenced by the acquisition of anchorage independent growth and tumorigenicity when injected into immunodeficient mice. Minor subpopulation of these cells (SAI) had the ability to migrate through soft agar and showed marked differences in cell morphology, proliferation, motility and expression of signaling proteins as well as higher tumorigenic potential. In addition, we examined changes between released extracellular vesicles (EV) from stem-like liver epithelial cell line WB-F344 (WB), their chemically transformed GP7TB and GP7TB.SAI, a soft agar invasive subpopulation. Apart from changes in number, size and proteomic content of EV, we discovered that GP7TB.SAI derived EVs significantly stimulated NK-mediated cytotoxicity opposite to those purified from the GP7TB line, despite their higher expression of Necl-5.
