Abstract
Proteini s domenama MATH i BTB (MAB) su od iznimne važnosti u embrionalnom razvoju mnogih vrsta. Upleteni su i u uspostavljanje reda mreže kortikalnih mikrotubularnih niti koje izgrađuju vreteno tijekom stanične diobe. Pored toga, oni su dio kompleksa kulin E3 ligaze, kojoj povećavaju specifičnost prema različitim supstratima i sudjeluju u njihovoj razgradnji. Cilj ovog rada bio je identificirati gene MAB u genomu bundeve C. pepo. Služeći se slijedom gena BPM1 iz vrste A. thaliana, konstruirane su probe za hibridizaciju po Southernu pomoću kojih je dokazana postojanost homolognih gena u vrste C. pepo. Na temelju sličnosti sekvenci gena s MATH i BTB domenama iz različitih biljnih vrsta konstruirane su degenerirane početnice koje su upotrijebljene za umnažanje gena u bundevi. Degeneriranim početnicama na kalupu genomske DNA bundeve umnožene su međugenske razmaknice ribosomalnih gena, dok na kalupu komplementarne DNA embriogenog tkiva bundeve nije bilo moguće umnožiti gene MAB.
Abstract (english)
MATH-BTB domain-containing proteins (MAB) are essentially important in a variety of species during the time of embryogenesis and post-embryonic development. Also, MAB proteins are involved in establishing the order within the cortical microtubule network which builds the spindle during the cell division, and increase the specificity of cullin E3 ligase against different substrates. Because of its importance, the aim of this study was to investigate the existence of genes with the MATH and BTB domains in the C. pepo genome. Using the gene sequence of the BPM1 gene from A. thaliana, the probes for the Southern hybridization were constructed and MAB genes were identified in C. pepo. Based on similarities between different MAB gene sequences, primers were constructed for amplification of homologuos genes in pumpkin. Using genomic DNA as template, intergenic spacers of ribosomal genes were amplified, and using complementary DNA from embriogenic pumpkin tissue as template, no gene was amplified.
