Abstract
Područje rijeke Save značajno je zbog svoje guste naseljenosti i izvora pitke vode za milijune ljudi, te je njena zaštita od velikog gospodarskog i ekološkog interesa. U sklopu projekta NATO SfP pod nazivom Assessment of Hazardous chemical contamination in the Sava River basin, proveden je suvremen pristup procjene rizika onečišćenja rijeke Save, tzv. EDA (eng. – Effects Directed Analysis). EDA – pristup, za razliku od uobičajenih kemijskih analiza, pruža svrsishodniji fokus na pojedine grupe zagađivala koristeći rezultate bioloških testova. Kao test utvrđivanja estrogenog potencijala u okolišnim uzorcima rijeke Save, korišten je biotest YES (eng. – yeast estrogen screen) zbog pouzdane mogućnosti utvrđivanja estrogenosti. Testni organizam je modificiran soj pekarskog kvasca Saccharomyces cerevisiae koji induciran estrogenom eksprimira enzimsku aktivnost β-galaktozidaze. Rezultati analize površinske vode i sedimenta rijeke Save biotestom YES ukazali su na umjereno zagađenje ksenoestrogenima, s nešto povišenim vrijednostima nizvodno od Zagreba, Siska i Jasenovca, dok razlog za zabrinutost pokazuju visoke vrijednosti dobivene za uzorak otpadne vode primarnog ispusta uređaja za preradu otpadnih voda grada Zagreba, ponovo ukazujući na nužnost prerade otpadnih voda.
Abstract (english)
Due to its drinking water reservoirs and dense population the River Sava basin represents the area of economic and environmental interest. Establishing a system for scientifically based protection, monitoring and risk assessment, a state of the art approach called the Effects Directed Analysis (EDA) was applied in this study. EDA combines results of the advanced chemical analysis and bioassays to identify the most critical categories of pollutants. In order to make assessment of estrogenic potential of the surface and sediment samples from the Sava River, the Yeast Estrogen Screen (YES) was used due to its simplicity, sensitivity and reliability. The test organism was a strain of genetically modified yeast, Saccharomyces cerevisiae, which expresses enzyme activity of β-galactosidase when induced by estrogens. Results indicated moderately elevated pollution by xenoestrogens downstream from Zagreb, Sisak and Jasenovac. As expected, results obtained by testing the primary effluent samples from the Zagreb city sewage treatment plant revealed high content of xenoestrogens, justifying an advanced waste water treatment of highly polluted waste waters of the Zagreb city.
