Abstract
Imazalil, cipermetrin i karbendazim su detektirani u biljkama za ljudsku prehranu. Kako bi istražili da li njihove kombinacije, primijenjivane oralno u niskim dozama, izazivaju imunotoksičnost, proveden je in vivo eksperiment. Doze od 10 mg/kg imazalila i cipermetrina te 20 mg/kg karbendazima i njihove kombinacije davane su miševima dnevno tijekom 28 dana. Nakon 24 sata od zadnje doze, biomarkeri oksidativnog stresa su mjereni u imunološkim organima: aktivnost katalaze i superoksid dismutaze te koncentracije reduciranog glutationa, malonildialdehida i 8-iso prostaglandina F2α. U usporedbi s kontrolnom skupinom, ovi markeri su promijenili aktivnost i koncentracije. Naši rezultati sugeriraju da pesticidi imaju potencijal izazvati oksidativni stres u imunološkim organima. Smanjenje koncentracija reduciranog glutationa utvrđene su u svim skupinama, osim kod skupine izložene karbendazimu u timusu i limfnim čvorovima. Opće povećanje koncentracije malonildialdehida je zabilježeno u svim skupinama, što ukazuje na to da se toksični učinak ovih pesticida vjerojatno vrši putem stvaranja slobodnih radikala. Povećanje aktivnosti superoksid dismutaze i katalaze u slezeni ukazuje na prekomjernu proizvodnju reaktivnih kisikovih vrsta.
Abstract (english)
Imazalil, cypermethrin and carbendazim are detected in plants for human nutrition. To explore whether their combinations, applied orally in low doses, would induce immunotoxicity, a subchronic in vivo experiment was conducted. Doses of 10 mg/kg of imazalil and cypermetrin and 20 mg/kg of carbendazim and their combinations were given to Swiss mice daily during 28 days. After 24 hours from the last dose, oxidative stress biomarkers in immune system organs were analysed: catalase (CAT) and superoxide dismutase (SOD) activities, malonilaldehyde (MDA), 8-iso prostaglandin F2α and glutathione (GSH) concentrations. Compared to control group these markers changed their activities and concentrations. Our results suggested that these pesticides have the potential to induce oxidative stress of the immune system. Decreases in GSH concentrations were determined in all groups except for groups with carbendazim in thymus and lymph node. A general increase in malonilaldehyde (MDA) concentration was observed in all test groups, which suggested that the toxic effects of these pesticides were probably exerted through free radical generation. Increases in superoxide dismutase and catalase activities in spleen point to the excessive production of reactive oxygen species.
