Abstract
Patogeni mikroorganizam Aphanomyces astaci (Oomycetes) uzročnik je račje kuge koji se preko invazivnih sjevernoameričkih slatkovodnih rakova proširio Europom. Sjevernoameričke vrste su zahvaljujući koevoluciji s patogenom razvile otpornost na razvoj bolestii te djeluju kao vektori širenja zaraze. S druge strane, europske autohtone vrste izrazito su podložne zarazi te, ukoliko dođe do kontakta s patogenom, dolazi do razvoja smrtonosne bolesti. Račja kuga jedan je od glavnih uzroka smanjenja populacija autohtonih vrsta rakova te je od velike važnosti pratiti njezinu prisutnost i širenje. S obzirom da postojeće metode detekcije A. astaci uključuju usmrćivanje rakova, cilj ovog rada bio je razviti neinvazivnu metodu detekcije patogena te tako izbjeći žrtvovanje ugroženih autohtonih vrsta. Temelj metode je uzorkovanje površinske mikrobiote trljanjem kutikule živog raka. Iz tako dobivene mješovite epibiontske mikrobne zajednice izolira se ukupna DNA te se PCR-testom provjerava da li je u uzorku prisutna DNA uzročnika račje kuge. U sklopu razvoja metode povećana je osjetljivosti PCR-testa pa je bilo moguće detektirati vrlo male količine DNA patogena. Rezultati novorazvijene metode podudarali su se s rezultatima tradicionalnog invazivnog načina detekcije. Novorazvijena metoda u budućnosti može služiti u razvijanju metoda liječenja ili prevencije račje kuge.
Abstract (english)
The pathogen Aphanomyces astaci (Oomycetes) is the causative agent of the crayfish plague which has expanded through Europe via invasive North American crayfish. Alien crayfish coevolved with the pathogen and are generally resistant to the disease. Contrary, native crayfish species are highly susceptible and infestation with A. astaci causes lethal disease. Crayfish plague has caused major declines of native european freshwater crayfish, thus monitoring of A. astaci presence and expansion is of paramount importance. Current methods for detection of A. astaci are based on tissue sampling for which animals have to be sacrified. Thus the main goal of this research was to develop a non-invasive method for A. astaci detection and avoid sacrifying endangered native crayfish. The method is based on collecting mixed epibiotic microbial communities, including A. astaci, by scrubbing the crayfish. Then, the DNA is isolated from mixed epibiotic microbial communities and the presence of A. astaci DNA in the sample is detected by PCR. The sensitivity of PCR was improved to detect very low amounts of A. astaci DNA. The efficacy of the newly developed non-invasive method was confirmed by comparison with tradional invasive method. This non-invasive approach could be used for treatment and monitoring of crayfish plague in the future.
