Abstract
Cilj rada je bio istražiti učinke resveratrola i njegovih nanokristala u dozi od 25 mg/kg na inhibiciju rasta Ehrlichovog ascitesnog tumora (EAT), broj krvnih žila u peritoneumu i mehanizam stanične smrti, te istražiti odnos između razine oštećenja jetre i bubrega kroz histološku analizu prisutnosti stanica u mitozi, apoptozi i nekrozi te razine oksidacijskog stresa u njima. Tumor smo prouzročili intraperitonealnim (ip) injiciranjem 2,5 × 106 EAT stanica u trbušnu šupljinu miševa soja Swiss albino. Obrada životinja s EAT tumorom po skupinama započeta je idućeg dana ip injiciranjem resveratrola u dozi od 25 mg/kg i nanokristala resveratrola u dozi od 25 mg/kg svaki drugi dan u periodu od 14 dana. Kontrolna skupina obrađena je ip s fiziološkom otopinom. Rezultati ukazuju da resveratrol i nanokristali resveratrola imaju inhibicijski učinak na rast stanica tumora, antioksidacijski i zaštitni učinak te zaštitnu ulogu u očuvanju fiziološke funkcije bubrega i jetre. Mikroskopska analiza broja krvnih žila u peritonealnoj ovojnici potvrđuje veliki antiangiogeni potencijal resveratrola i njegovih nanokristala što posljedično vodi inhibiciji rasta tumora.
Abstract (english)
The aim of the study was to investigate the effects of resveratrol and its nanocrystals at a dose of 25 mg/kg on inhibition of Ehrlich ascites tumor growth, blood count in the peritoneum and mechanism of cell death, and to investigate the relationship between liver and kidney damage levels through histological analysis of the presence of cells in mitosis, apoptosis and necrosis, and levels of oxidative stress in them. The tumor was caused by intraperitoneal (ip) injection of 2.5 x 106 cells into the abdominal cavity of Swiss albino mice. Treatment of animals with EAT tumor in groups was started the next day by injecting resveratrol at dose of 25 mg/kg and resveratrol nanocrystals at dose of 25 mg/kg every other day for 14 days. The control group was treated ip with saline. The results indicate that resveratrol and resvertrol nanocrystals have an inhibitory effect on tumor cell growth, an antioxidant and protective effect, and a protective role in preserving the physiological function of the kidney and liver. Microscopic analysis of the number of blood vessels in the peritoneal sheath confirms the great antiangiogenic potential of resveratrol and its nanocrystals, which in turn leads to inhibition of tumor growth.
