Abstract
Nanočestice srebra (AgNP) jedan su od najčešće korištenih nanomaterijala. Usprkos širokoj primjeni, njihov utjecaj na žive organizme je i dalje predmet istraživanja. U ovom radu je istražen utjecaj triju vrsta nanočestica srebra, stabiliziranih različitim omotačima, na vijabilnost i oksidativni stres ljudskih keratinocita. Pri tome se istražio učinak AgNP u i bez prisustva proteina koji u biološkim sustavima tvore na nanopovršini proteinsku koronu. Proteinska korona može modulirati interakciju nanočestica sa živim sustavima. Rezultati dobiveni primjenom MTS metode na mikrotitarskom čitaču i metode Live/Dead na protočnom citometru pokazuju da su negativno nabijene AgNP citotoksične pri najvišoj testiranoj koncentraciji u uvjetima bez proteinske korone, dok su pozitivno nabijene AgNP pokazale citotoksičnost u prisustvu proteinske korone. Neutralne AgNP nisu pokazale citotoksičnost u ispitivanom koncentracijskom rasponu. Mjerenjem ukupnog sadržaja slobodnih radikala i glutationa (GSH) nije dokazan oksidativni stres u ljudskih keratinocita nakon tretmana nanočesticama srebra.
Abstract (english)
Silver nanoparticles (AgNP) are one of the most widely used types of nanomaterials. However, their effects on living organisms is still the subject of ongoing research. In this thesis, cell viability and oxidative stress response of human keratinocytes exposed to three different types of AgNP were investigated. In addition, cell response was evaluated after addition of proteins in test media to determine the effect of protein corona formed on AgNP surface. Recent studies suggest that protein corona may modulate the biological effect of nanoparticles. Testings were performed by MTS assay and Live/Dead flow cytometry tests. Obtained results demonstrated that negatively charged AgNP were cytotoxic to keratinocytes only at the highest concentrations applied without the presence of protein corona, while positively charged AgNP showed cytoxicity at the highest tested concentrations after protein corona formation. Neutral AgNP were not cytotoxic in the tested concentration range. Oxidative stress response in cells was tested by mesuring total amount of peroxy radicals and glutation. Results did not indicate any oxidative stress in human keratinocytes exposed to tested concentrations of AgNP.
