Abstract
Fitoplazme ('Candidatus Phytoplasma') su bakterije bez stanične stijenke koje koloniziraju biljke i neke skupine kukaca. Genom fitoplazmi sadrži mnogo ponavljajućih jedinica te ponekad i izvankromosomske elemente na kojima se mogu nalaziti geni prisutni u više kopija u genomu. Jedan od tih gena je i gen za proteine SSB. Proteini SSB imaju esencijalnu ulogu u preživljavanju stanice i nalaze se u svim domenama živog svijeta, ali i virusa. Geni za proteine SSB prisutni su u različitom broju u genomima fitoplazmi, no postoji samo jedna kopija koja ima drugačije evolucijsko porijeklo od ostalih te se smatra izvornom kopijom gena ssb. Izvorna kopija gena ssb većeg broja fitoplazmatskih izolata vrsta 'Ca. P asteris' i 'Ca. P. solani' s područja Hrvatske umnožena je metodom PCR i sekvencirana. Filogenetskom analizom pokazana je očuvanost izvorne kopije gena ssb. Nadalje, potvrđeno je da postoji jedna kopija tog gena u genomu izolata fitoplazmi vrste 'Ca. P. asteris', dok je kod biljnog uzorka vrste 'Ca. P. solani' dokazana još jedna kopija tog gena, koja se ne pojavljuje kod uzorka fitoplazmi izoliranog iz kukca.
Abstract (english)
Phytoplasmas (‘Candidatus Phytoplasma’) are bacteria without cell wall that colonize plants and some groups of insects. They have repeat rich genomes with extrachromosomal elements that are mainly composed of multicopy genes. One of those multicopy genes is ssb gene. SSB proteins, essential for cell survival, are found in all domains of life as well as in viruses. In phytoplasma genomes, SSB protein genes may be present in different numbers but there is only one original copy of ssb gene that has different evolutionary origin from the others. Original copy of ssb gene from a number of 'Ca. P. asteris' and 'Ca. P. solani' isolates originated from different areas of Croatia was amplified by PCR and sequenced. Phylogenetic analysis of ssb gene sequences has shown that original copy of ssb gene is conserved. In addition, one copy ssb gene was confirmed in the genomes of ‘Ca. P. asteris’ isolates, whilst there is another copy present in the plant isolates of ‘Ca. P. solani’ that isn’t present in genome isolate of ‘Ca. P. solani’ obtained from insect vector.
