Abstract
Šećerna bolest ili dijabetes (lat. diabetes mellitus; DM) je kronični poremećaj metabolizma ugljikohidrata, masti i bjelančevina uzrokovan relativnim ili apsolutnim nedostatkom inzulina, s posljedičnom hiperglikemijom. Cilj ovog istraživanja bio je proučiti moguće zaštitne učinke naringenina na nastala oštećenja u miševa s dijabetesom prouzročenim aloksanom. Tip 1 dijabetesa induciran je u miševa davanjem aloksana u dozi od 75 mg kg-1 intravenozno. Nakon 2 dana dolazi do povećanja glukoze u serumu životinja obrađenih aloksanom a 10. dana od iniciranja aloksana vidljiv je znatni pad tjelesne mase u odnosu na zdravu kontrolnu skupinu. Naringenin (50 mg kg-1) iniciran miševima s dijabetesom tijekom sedam dana bio je učinkovit u održavanju normalne tjelesne mase životinja, smanjenju histopatoliških promjena u jetri i bubregu te povećanju funkcionalne aktivnosti makrofaga peritonealne šupljine i preživljenju životinja. Rezultati komet testa ukazuju da je naringenin smanjio oštećenja DNA na limfocitima periferne krvi uzrokovana oksidativnim stresom te da naringenin ima obećavajući učinak na smanjenje negativnih posljedica razvoja dijabetesa, čemu doprinosi njegov antioksidativni potencijal.
Abstract (english)
The present study was designed to investigate the possible antidiabetic and protective effect of naringenin against alloxan-induced oxidative damage in diabetic mice. Type 1 diabetes was induced with alloxan in dose of 75 mg kg-1 btw iv. After 2 days appears to increase glucose levels in alloxan treated animals, and 10th days of the initiation of alloxan show significant decrease of body weight compared to a healthy control group. Treatment of diabetic mice with naringenin in dose of 50 mg kg-1 for seven days reduced the decline in body mass, reduced histopathologycal changes of liver and kidney, increased functional activity of macrophages peritoneal cavity and increases the survival of mice. Additionaly comet assay results show that treatment with naringenin reduces the alloxan-induced DNA damage to lymphocytes. The results indicate that naringenin has promising effects in reducing the negative consequences of developing diabetes, which contributes to its antioxidant potential.
