Abstract
Neobične (ili rijetko korištene) aminokiseline su prisutne u nekolicini staničnih peptida. Često su uključene u peptidne antibiotike proizvedene ne-ribosomalnom sintezom, a u drugim slučajevima do njihove ribosomalne ugradnje dolazi prilikom ekspresije heterolognih proteina u bakterijskim stanicama. Norvalin je jedna od takvih, nekanonskih aminokiselina koji može biti ugrađen u proteine na leucinske pozicije. Razvijene su i metode koje omogućuju ciljanu ugradnju nekanonskih aminokiselina u proteine te tako povećavaju strukturalnu različitost proteinskih bočnih ogranaka. Proširivanjem seta od 20 aminokiselina uporabom nekanonskih analoga, omogućeno je sustavnije istraživanje različitih fizikalno-kemijskih fenomena. Osim korištenja u fundamentalnim istraživanjima, veliki je interes i za ugradnju neproteinogenih aminokiselina u prirodne spojeve i peptide za terapeutsku uporabu kako bi se poboljšala njihova svojstva i učinkovitost. Korištenjem ovakvog proširenog genetičkog koda pružaju se mogućnosti za stvaranje modelnih proteina te proteina s poboljšanim ili potpuno novim svojstvima.
Abstract (english)
Unusual (or infrequently occurring) amino acids are present in a number of cellular peptides. They are often included in peptide antibiotics produced by nonribosomal synthesis, in other cases, misincorporation of these amino acids into proteins by the ribosomes occurs when heterologous proteins are expressed in bacterial cells. Norvaline is one of those noncanonical amino acids that can be incorporated in proteins on leucine positions. Methods that allow site-specific incorporation of noncanonical amino acids were developed; thereby increasing the structural diversity of amino acid residues used in proteins. By expanding the natural ocurring set of 20 proteinogenic amino acids, more systematic research of different physicochemical phenomena is enabled. Except for its use in fundamental research, there is a great interest in incorporation of nonproteinogenic amino acids in natural products and peptides for therapeutic use. Usage of this expanded genetic code offers possibilities for designing model peptides and proteins with enhanced or novel properties.
