Abstract
Autofagija je proces uključen u membranski prijenos proteina, organela, patogena i različitih nakupina do lizosoma, za razgradnju. Autofagiju čini široki molekularni sustav reguliran brojnim molekularnim mehanizmima. Proces makroautofagije zahtijeva nastajanje dvostruko-membranskog sloja koji okružuje ciljani materijal. Sloj dvostruke membrane se zatvara, te se s vremenom spaja s lizosomom. Jedan od glavnih regulatornih proteina u autofagiji je “mammalian target of rapamycin”, odnosno mTOR. Ranija istraživanja su pokazala kako je protein “Pleckstrin homology domain-containing family M member 1”, kraće Plekhm1, uključen u autofagiju. Mutacija u genu Plekhm1 vodi do osteopetroze, bolesti koja utječe na resorpciju kostiju. Naši neobjavljeni rezultati pokazuju da bi Plekhm1 mogao biti povezan uz kasnije stadije autofagije, te da reagira s proteinima Rab7 i LC3, nekima od pokazatelja kasnijih stadija autofagije. U ovom radu pokušavamo dati kratak uvid u regulaciju Plekhm1 u autofagiji i interakciju istog proteina s različitim markerima autofagije.
Abstract (english)
Autophagy is a process involved in membrane trafficking of proteins, organelles, pathogens and aggregates to lysosomes for degradation. Autophagy consists of large molecular machinery, regulated by numerous mechanisms. The process of macroautophagy requires the de novo formation of a double membrane layer that surrounds the target material, seals and eventually fuses with the lysosome. One of the key regulators in autophagy is mammalian target of rapamycin, mTOR. Previous studies showed the involvement of a protein called Pleckstrin homology domain-containing family M member 1, Plekhm1 in osteopetrosis, a disease affecting bone resorbtion. Our unpublished data suggests that Plekhm1 is involved in later stages of autophagy. Also, our data suggests that Plekhm1 interacts with Rab7 and LC3 proteins, which are indicatiors of late phase autophagy. Here, we give a brief insight into Plekhm1 regulation and highlight novel interactions with proteins involved in autophagy.
