Abstract
Vodeći uzrok smrtnosti u svijetu su kardiovaskularne bolesti. Uzrokovane su povećanom koncentracijom lipoproteina, čijim gomilanjem na meandrima krvnih žila nastaje aterosklerozni plak te uzrokuje perpetuirano ometanje protoka krvi. Bolesnici koji pate od hiperlipoproteinemije konzumiraju lijekove tzv. statine, koji inhibiraju djelovanje enzima 3- hidroksi-3-metilglutarilkoenzim A-reduktaze potrebnog za sintezu kolesterola. U konačnici, statini uzrokuju uklanjanje viška štetnih lipoproteina u krvi. Rosuvastatin uzrokuje manje nuspojava od ostalih lijekova skupine statina te ima pleiotropno djelovanje neovisno o primarnoj svrsi djelovanja. Svrha ovog rada bila je ispitati razgradne produkte rosuvastatina i njegovu strukturnu stabilnost nakon ozračivanja uzorka gama zrakama pomoću 60Co izvora zračenja. Identifikacija produkata provedena je nanoUPLC-ESI-MS/MS sustavom, kojim je određen elementni sastav šest razgradnih produkata rosuvastatina.
Abstract (english)
Cardiovascular diseases are one of the leading causes of death globally. They usually arise when high levels of lipoproteins in meanders of blood vessels form an atherosclerotic plaque which then obstructs blood circulation. Patients diagnosed with hyperlipoproteinemia use drugs called statins that inhibit 3-hydroxy-3-methyl glutaryl coenzyme A reductase required for cholesterol synthesis. Ultimately, statins eliminate adverse lipoproteins from blood. Rosuvastatin causes less side effects than other drugs in the statin group and possesses pleiotropic effects independent of its primary use. The purpose of this study was to investigate rosuvastatin degradation products and structural stability when exposed to gamma radiation using 60Co. Subsequent analysis was performed by nanoUPLC-ESI-MS/MS which determined elemental composition of six degradation products of rosuvastatin.
