Abstract
Psorijaza je kronična, upalna, proliferativna bolest kože čije su glavne značajke brzo razmnožavanje stanica, diferencijacija, nemogućnost proizvodnje keratina, pojava crvenila i deskvamacija na kojima se temelji histopatološka analiza epidermisa. Cilj ovog rada bio je primijeniti infracrvenu kameru koja bilježi promjene tjelesne temperature i razvoj upale na animalnom modelu psorijaze, te temeljem istih parametra zabilježiti antioksidativnu i antiupalnu učinkovitost kvercetina. U ispitivanju ovih problema, poslužili su eksperimenti koji su uključivali patohistološku i biokemijsku analizu, praćenje parametra oksidativnog stresa kao i primjenu infracrvene kamere. Ukupan broj upalnih stanica, kao i diferencijalna analiza stanica pokazuju smanjenje broja stanica osobito limfocita čime je dokazano antiproliferativno i antiupalno djelovanje kvercetina. Pozitivan učinak kvercetina nije se samo odnosio na upalni stanični infiltrat nego i na znatno manji indeks širenja makrofaga u trbušnoj šupljini, te je dokazana apoptotička aktivnost kvercetina. Isto tako, praćenjem biljega oksidativnog stresa u psorijazi, kvercetin je pokazao izrazito antioksidativno djelovanje.
Abstract (english)
Psoriasis is a chronic, inflammatory, proliferative skin disease whose main features are rapid cell proliferation, differentiation, lack of keratin production, redness and desquamation all on which the histopathological analysis of the epidermis is based on. The aim of this study was to apply an infrared camera that records the changes in body temperature and the development of inflammation in animal models of psoriasis, and based on these parameters record the antioxidant and anti-inflammatory effects of quercetin. Experiments involving histopathological and biochemical analysis, monitoring oxidative stress parameters and the application of infrared cameras. The total numbers of inflammatory cells, as well as the differential analysis of the cells both showed a decrease in cell numbers distinctively of lymphocytes, which proved the antiproliferative and anti-inflammatory properties of quercetin. The positive effect of quercetin is not only related to inflammatory cell infiltration but also a much lower spreading index of macrophages in the abdominal cavity, and the explicit apoptotic activity of quercetin. The monitoring of oxidative stress markers in psoriasis also demonstrated the clear antioxidant activity of quercetin.
