Abstract
Šikonin, prirodni spoj izoliran iz korijena biljke Lithospermum erythrorhizon Sieb. et Zucc., od davnina se koristi u tradicionalnoj kineskoj medicini za liječenje dermatitisa, rana, ospica i opeklina, a danas je poznat po svojem protuupalnom, antioksidativnom i protutumorskom djelovanju. U ovom radu ispitivano je protutumorsko djelovanje šikonina na stanice tumora mokraćnog mjehura. Ispitivan je njegov utjecaj na stanično preživljenje ovisno o koncentraciji, gustoći nasijanih stanica i volumenu u kojem su stanice tretirane, a indukcija određenog oblika stanične smrti ispitivana je mjerenjem ekspresije proteina i gena karakterističnih za apoptozu i nekroptozu. S obzirom da apoptoza i nekroptoza mogu biti inducirane preko TNFα/TNFR signalnog puta, pomoću inhibitora apoptoze (ZVAD) i inhibitora nekroptoze (Nec1s), ispitivano je može li se određena stanična smrt inhibirati i preusmjeriti iz jednog oblika u drugi. Rezultati su pokazali da šikonin uspješno djeluje na tumorske stanice mokraćnog mjehura inhibirajući staničnu proliferaciju te inducira i apoptozu i nekroptozu. Uspješno preusmjerenje stanične smrti pomoću tretmana šikoninom može biti ključno u prevenciji tumora rezistentnih na kemoterapeutike koji induciraju apoptozu.
Abstract (english)
Shikonin, a natural compound isolated from the roots of the plant Lithospermum erythrorhizon Sieb. et Zucc., has long been used in traditional Chinese medicine to treat dermatitis, wounds, measles and burns, and today is known for its anti-inflammatory, antioxidant and antitumor effects. In this thesis the antitumor effect of shikonin on bladder cancer cells was investigated. How it effects cell survival depending on the concentration, density of seeded cells and the volume in which the cells were treated and the induction of a different forms of cell death which was examined by measuring the expression of proteins and genes characteristic for apoptosis and necroptosis. Since apoptosis and necroptosis can both be induced trough the TNFα / TNFR signaling pathway, apoptosis inhibitors (ZVAD) and necroptosis inhibitors (Nec1s) have been investigated to determine whether a particular cell death can be inhibited and redirected from one form to another. The results showed that shikonin successfully acts on bladder cancer cells by inhibiting cell proliferation and inducing both apoptosis and necroptosis. Successful redirection of cell death by shikonin treatment may be crucial in the prevention of tumors resistant to other apoptosis-inducing chemotherapeutics.
