Abstract
T-stanični limfomi čine 10% ne-Hodgkinovih limfoma te najčešće nastaju iz zrelih T-stanica timusa čime pripadaju skupini perifernih limfoma T-stanica. Limfomi ove skupine su zbog rijetke zastupljenosti slabije istraženi. Cilj ovog rada je utvrditi potencijalnu promjenu ekspresije transkripcijskog faktora BACH2 u tumorskim stanicama perifernog T-staničnog limfoma bez specifičnih obilježja. Stanice T-zone netumorskog sekundarnog limfnog tkiva izolirane su pomoću mikrodisektora i korištene kao kontrola. Izolirana RNA je prevedena u cDNA metodom reverzne transkripcije te je ekspresija gena BACH2 i endogene kontrole, gena HPRT1, analizirana metodom qPCR. Korištenjem relativne kvantifikacije ekspresije gena utvrđeno je značajno smanjenje ekspresije BACH2 u tumorskom tkivu. Smanjenje ekspresije upućuje na promjene u tumorskim stanicama koje pridonose mehanizmu izbjegavanja apoptoze. Ovakvi rezultati suprotni su dostupnim podacima o promjenama ekspresije BACH2 u većini tipova B- staničnih limfoma gdje je njegova prekomjerna ekspresija povezana s lošijom prognozom oboljelih što upućuje na drugačiju ulogu ovog transkripcijskog faktora ovisno o unutarstaničnom miljeu.
Abstract (english)
T-cell lymphomas make 10% of non-Hodgkin lymphomas and most often develop from mature T-cells in thymus which makes them part of peripheral T-cell lymphoma subtype. Lymphomas of this subtype are poorly understood because of their rare occurrence. The aim of this research is to determine the potential change in expression of transcription factor BACH2 in tumour cells of peripheral T-cell lymphoma, not otherwise specified. T-cell zone from histological specimens of healthy secondary lymphoid tissue was separated with microdisector and used as a control. Isolated RNA was reverse-transcribed into cDNA and the expression of BACH2 was quantified by qPCR, with HPRT1 gene used as endogenous control. Relative quantification of gene expression showed significantly lower expression of BACH2 in tumour tissue. Lower expression indicates changes in tumour cells that contribute to mechanisms of apoptosis avoidance. The results are different from the observations of BACH2 expression in B-cell lymphoma where its lower expression indicates poorer survival which points out to different roles of this transcription factor depending on the inside of the cell.
