Abstract
Alzheimerova bolest (AB) je neurodegenerativna bolest u kojoj propadaju neuroni kortikalnih i subkortikalnih regija mozga. Neurodegeneracija zahvaća neurone koji sintetiziraju neurotransmitere kao što je serotonin. Hipoteza rada je bila da su varijante gena za serotoninski receptor tipa 2C (5-HT2C) i za apolipoprotein E (APOE) rizični čimbenici za razvoj AB. Cilj rada je bio odrediti polimorfizme gena za 5-HT2C (rs3813929; -759C/T) i APOE (rs7412 i rs429358) u 366 zdravih osoba starije dobi i 207 bolesnika s AB. Rezultati su pokazali: a) da između oboljelih osoba i zdrave kontrole nema razlike u distribuciji genotipova i alela polimorfizma -759C/T gena za 5-HT2C, b) da je frekvencija alela E4 gena za APOE značajno viša u oboljelih od AB i c) da nema sinergističkog učinka ovih dvaju gena. Rezultati su potvrdili da je alel APOE4 rizični čimbenik za nastanak AB, ali nisu potvrdili pretpostavku o povezanosti polimorfizma gena za 5-HT2C i AB.
Abstract (english)
Alzheimer's disease (AD) is a neurodegenerative disorder characterized by degeneration of the neurons in cortical and subcortical brain regions. Neurodegeneration affects the neurons that synthesize neurotransmitters like serotonin. Our hypothesis was that genetic variants of serotonergic receptor type 2C (5-HT2C) and apolipoprotein E (APOE) are risk factors for the development of AD. The aim of the study was to determine polymorphism (rs3813929; -759C/T) of 5-HT2C gene and APOE (rs7412 and rs429358) gene in 366 healthy elderly persons and 207 patients with AD. Results showed: a) no difference in genotype and allele frequencies of -759C/T 5-HT2C gene polymorphism between patients and healthy controls, b) the significant increase in allele E4 frequency in patients with AD, c) no sinergistic effect of these genes. Our results confirm that alele E4 could be a risk factor for AD, but do not support the hypothesis that polymorphism of 5-HT2C receptor gene is related to AD.
