Abstract
Hydrophilidae (vodoljupci) je brojem vrsta velika porodica vodenih kornjaša čije predstavnike karakterizira morfološka raznolikost i prilagođenost na različite tipove staništa. Kako su Hydrophilidae slabo istražena skupina u Hrvatskoj, terenska istraživanja su provedena u njenim različitim područjima u razdoblju od 29.05. 2010 do 25.08. 2011. godine. Glavni cilj istraživanja je razjasniti srodstvene odnose unutar porodice Hydrophilidae koji bi pomogli u reviziji kroz koju ta porodica prolazi. Uzorci su determinirani prema morfološkim obilježjima te je iz njih izolirana DNA koja je kasnije korištena u filogenetskim analizama pomoću dva molekularna biljega: mitohondrijski geni za citokrom oksidazu podjedinica I (COI) i za manju podjedinicu ribosomalne RNA (12S rRNA). Rezultati filogenetskog istraživanja pokazuju da COI i 12S rRNA daju dobru razlučivost na nižim razinama (vrste i rodovi) te da su prikladni za analize na toj razini, ali nisu dovoljno informativni za analize dubljih filogenetskih odnosa na višim granama (podporodice, porodice), te da su analize pouzdanije ako se vrše na združenim molekularnim biljezima (COI i 12S rRNA). Ovo istraživanje je dalo temelje za buduća, kako taksonomska tako i filogenetska istraživanja na porodici Hydrophilidae Hrvatske i Europe.
Abstract (english)
Family Hydrophilidae is large family of water beetles with morphologically diverse representatives that live in various habitats. According to the fact that hydrophilida are poorly investigated in Croatia, field research was carried out in different areas of country in the period from May 29th, 2010-August 25th, 2011. The main objective of this study was to examine and clarify evolutionary relationships within the family Hydrophilidae, expecting that it would help in revisions this family is currently undergoing. Samples were determined using morphological characteristics. DNA was isolated from the samples and phylogenetic analyses were conducted by the use of two mitochondrial markers: genes for cytochrome oxidase subunit 1 (COI) and for smaller subunit of ribosomal RNA (12S rRNA). The results of research indicate that COI and 12S rRNA provide good distinguishing features on lower levels (species and genus) and that they are suitable for analyses on that level, but not sufficiently informative for analyses of deeper phylogenetic reltionships (on the subfamily and family level). Additionally, it has been found that analyses are more reliable if they are conducted with joined molecular markers (COI and 12S rRNA). This research provides the basis for the future studies, both taxonomic and phylogenetic, on hydrophilids in Croatia and Europe.
