Abstract
DNA barcoding je metoda koja omogućava detaljno proučavanje i razumijevanje postojanja određenih vrsta. Primjena ove metode zahtijeva korištenje male univerzalne sekvence DNA molekule, tzv. početnice koja se dovoljno razlikuje za svaku vrstu, kako bi se jedinke mogle identificirati na toj taksonomskoj razini. Temelj metode je određivanje standardiziranog dijela mitohondrijskog gena za podjedinicu I citokrom oksidaze (COI) koja je duga oko 650 pb. Pri određivanju biološke kvalitete vode identificiraju se jedinke makrozoobentosa koje reagiraju na sve uvjete u vodi i svojim ponašanjem, životnim ciklusima i prilagodbama na trenutne uvjete daju informaciju o stanju okoliša u kojem žive.
Abstract (english)
DNA barcoding is a method which enables detailed study and understanding of existence of certain species. Application of this method requiers usage of small universal sequence of the DNA molecule, so called primer or marker, which differs enough for each species so that individuals of species could be identified on that taxonomic rank. Foundation of the method is a determination of standardised part of mitochondrial gene for subunit I of the cytochrome oxidase (COI) which is about 650 bp long. In determining biological quality of water the individuals of macrozoobenthos are identified and they respond to all water conditions and by their behavior, life cycle and adjusments on those conditions they give information about current state of the enviroment in which they live.
