Abstract
Proces fosforilacije jedan je od najčešćih metoda regulacije ciljnih proteina koja omogućuje normalnu staničnu komunikaciju te doprinosi održavanju homeostaze. Odvija se pomoću proteinskih kinaza koje izvršavaju bitne uloge u gotovo svim staničnim signalnim putevima, uključujući staničnu proliferaciju, diferencijaciju, migraciju, apoptozu te metabolizam. Promijenjena aktivnost većine proteinskih kinaza, s naglaskom na proteinske tirozin-kinaze, uočena je kod mnogih ljudskih bolesti, osobito tumorskih. Upravo zato, ova vrsta enzima postala je središnja meta farmaceutske industrije za razvoj selektivnih inhibitora koji bi se mogli koristiti u liječenju takvih oboljenja. U ovom radu obrađeno je djelovanje proteinskih kinaza, s naglaskom na tirozin-kinaze, njihova signalizacija i uloga u zdravim i malignim stanicama. Također su predstavljeni pojedini inhibitori tirozin-kinaza te njihova uloga u liječenju raznih bolesti.
Abstract (english)
The process of phosphorylation is one of the most common methods of regulating target proteins, as it allows normal cellular communication and contributes to the maintenance of homeostasis. It occurs with the help of protein kinases, which play important roles in most cellular signaling pathways, including cellular proliferation, differentiation, migration, apoptosis and metabolism. Altered activity of most protein kinases, with an emphasis on the protein tyrosine kinases, is observed in many human diseases, especially tumors. That is why this type of enzyme has become the main target of the pharmaceutical industry for the process of developing selective inhibitors, which could be used as treatment against such diseases. This thesis examines the impact of protein kinases, with the emphasis on the tyrosine kinases, their signaling and their role in both healthy and malignant cells. Individual inhibitors of tyrosine kinases and their role in the treatment of various diseases are also presented.
