Abstract
Stanična komunikacija signalnim molekulama QS (engl. Quorum sensing) važan je regulatorni mehanizam koji koriste mnoge bakterijske vrste za koordinaciju ekspresije gena i ponašanja kao odgovor na promjene u gustoći populacije. U kontekstu stvaranja biofilma, stanična komunikacija QS vrlo je složen i strogo reguliran proces koji uključuje proizvodnju i detekciju specifičnih signalnih molekula kao što su autoinduktori. Autoinduktor 2 (AI-2) je od posebne važnosti za bakterijsku komunikaciju i razvoj biofilma. Prisutnost gena stanične komunikacije putem AI-2 kemijskih signalnih molekula (engl. Lux quorum) u bakterije Vibrio gigantis, uključujući luxPQ, luxU i luxO je intrigantna jer su ti geni općenito uključeni u regulaciju bioluminiscencije u drugim vrstama roda Vibrio. Međutim, nepostojanje transkripcijskog aktivatora LuxR i operona luciferaze (luxCDABEGH), odgovornih za proizvodnju enzima luciferaze koji su kritični za bioluminiscenciju, sugerira jedinstvenu genetsku konfiguraciju vrste V. gigantis. Kombinirani rezultati PICRUSt analize i eksperimentalnog testa bioluminiscencije, kao i promatranje dinamike formiranja mikrokolonija i stanica roja, potvrđuju kako V. gigantis ima genetski potencijal za stvaranje biofilmova pomoću sustava QS, ali nema sposobnost bioluminiscencije. Ovo je empirijski dokaz kako V. gigantis koristi mehanizme QS različite od drugih bioluminiscentnih vrsta i ukazuje na različite strategije koje bakterije koriste kako bi koordinirali svoje ponašanje u biofilmovima.
Abstract (english)
Quorum sensing (QS) is an important regulatory mechanism used by many bacterial species to coordinate gene expression and behavior in response to changes in population density. In the context of biofilm formation, quorum sensing is a highly complex and tightly regulated process that involves the production and detection of specific signaling molecules such as autoinducer 2 (AI-2), which is of particular importance for bacterial communication and biofilm development. The presence of genes associated with the Lux quorum-sensing system in Vibrio gigantis, including luxPQ, luxU, and luxO, is intriguing because these genes are generally involved in the regulation of bioluminescence in other Vibrio species. However, the absence of the transcriptional activator LuxR and the luciferase operon (luxCDABEGH), which are responsible for the production of the enzyme luciferase that is critical for bioluminescence, suggests a unique genetic configuration in V. gigantis. The combined results of the PICRUSt analysis and the experimental bioluminescence assay, as well as the observation of the dynamics of microcolony and swarm cell formation, confirm that V. gigantis has the genetic potential to form biofilms through quorum sensing, but lacks the ability to bioluminesce. This is empirical evidence that V. gigantis uses QS mechanisms distinct from bioluminescent species and sheds light on the different strategies bacteria employ to coordinate their behavior in biofilms.
