Abstract
Ciklosporin A (CsA) dobiven iz gljive Tolypocladium inflatum koju odlikuje produkcija velikog broja sekundarnih metabolita, iznimno je bitan imunosupresivni lijek. Mehanizam biosinteze ovog spoja u Tolypocladium inflatum tek je nedavno pobliže opisan, no njegova struktura kao i mehanizam djelovanja podrobno su istraženi te se zna da se on zbog učinkovitog formiranja kompleksa s citosolnim proteinom ciklofilinom veže na kalcineurin te koči njegovu funkciju, čime se zaustavlja aktivacija T stanica u organizmu. Zahvaljujući ovom mehanizmu CsA je izuzetno efektivan imunosupresiv koji se u suvremenoj medicini koristi nakon transplantacije organa kako bi se smanjila mogućnost odbacivanja novog organa zbog imunološke reakcije. Brojna istraživanja dokazala su njegovu efektivnost i kod tretiranja raznih imunoloških bolesti kao što su reumatoidni artritis i lupus, a učinkovitost je pokazao i kod ublažavanja simptoma brojnih dermatoloških bolesti (psorijaze, atopijskog dermatitisa, lichen planusa).
Abstract (english)
Cyclosporin A (CsA), derived from the fungus Tolypocladium inflatum, which is characterized by the production of a large number of secondary metabolites, is an extremely important immunosuppressive drug. The biosynthetic mechanism of this compound in Tolypocladium inflatum has only recently been elucidated in detail. However, its structure and mechanism of action have been thoroughly researched. It is known that CsA effectively forms a complex with the cytosolic protein cyclophilin, binding to calcineurin and inhibiting its function, thereby preventing T-cell activation in the body. Due to this mechanism, CsA is an exceptionally effective immunosuppressant used in modern medicine to reduce the risk of organ rejection following transplantation due to the immune response. Numerous studies have proven its effectiveness in treating various immune-related diseases such as rheumatoid arthritis and lupus, and it has also shown efficacy in alleviating symptoms of numerous dermatological conditions (psoriasis, atopic dermatitis, lichen planus).
