Abstract
Coscinasterias tenuispina is a fissiparous starfish, capable of both sexual and asexual reproduction. When reproducing asexually, it splits in half, thus creating two separate individuals. Each of these will regenerate the missing half of the body, in a complex process which is not fully elucidated, resulting in two new individuals capable of further reproduction. The subject of this thesis is to explore the aging and regeneration pattern in these clonal animals, which represent an interesting model for aging studies. Also, it has been suggested that some populations of C. tenuispina can proliferate without sexual reproduction, through fission only. If this is the case, then these populations either do not age, or they are capable of reversing the damage accumulated with age, meaning that they are capable not only to regenerate but also to rejuvenate. To test these possibilities, several well-known biomarkers of aging such as activities of catalase, superoxide dismutase, glutathione and proteasome were measured in both regenerating and non-regenerating arms. The results of measurement of all four parameters show a tendency of their decreasing with length of arm. The actual age of these individuals is unknown, so length of arm is used as a parameter that most likely correlates with their chronological age. Therefore, these results suggest that these starfish do indeed accumulate age-related damage. In highly asymmetrical individuals, there is a significant difference in measured parameters between regenerating and non-regenerating arms, implying that regenerating arms are younger than their nonregenerating counterparts. This might indicate rejuvenation on the level of tissue. In this experiment it was not possible to demonstrate that rejuvenation additionally occurs on cellular level. However, the possibility that some starfish populations can survive only through asexual reproduction implies the capability of cellular rejuvenation, and this possibility should be studied further.

Coscinasterias tenuispina je vrsta morske zvijezde koja se može razmnožavati spolnim ili nespolnim putem (fisijom). Pri nespolnom razmnožavanju, tijelo se prepolovi te nastaju dva odvojena primjerka. Svaki će regenerirati dio tijela koji nedostaje, u kompleksnom procesu koji nije u potpunosti razjašnjen, što u konačnici rezultira dvjema novim jedinkama sposobnim za daljnje razmnožavanje. Tema ovog rada je istraživanje starenja i regeneracije kod tih klonalnih životinja, koje predstavljaju zanimljiv model za istraživanja procesa starenja. Također, postoje indicije da se neke populacije C. tenuispina održavaju bez spolnog razmnožavanja, isključivo fisijom. Ukoliko je to točno, takve populacije ili ne stare, ili imaju sposobnost poništenja oštećenja koja su uzrokovanja starenjem, što znači da ove životinje imaju sposobnost ne samo regeneracije nego i pomlađivanja. U svrhu ispitivanja tih mogućnosti, mjerili smo nekoliko poznatih biomarkera starenja, kao što su aktivnost katalaze, superoksid dismutaze, glutationa i proteasoma, u regenerirajućim i neregenerirajućim krakovima. Rezultati mjerenja pokazuju za sva četiri parametra tendenciju njihovog smanjivanja sa duljinom kraka. Stvarna dob primjeraka nije poznata, pa koristimo duljinu kraka kao parametar koji najvjerojatnije korelira s njihovom kronološkom dobi. Stoga, ti rezultati ukazuju da ispitivane jedinke doista nakupljaju oštećenja uzrokovana starenjem. Kod primjeraka koji su vrlo asimetrični, postoji značajna razlika u mjerenim parametrima između regenerirajućih i neregenerirajućih krakova, što upućuje na mlađi biološki status regenerirajućih krakova u odnosu na pripadajuće neregenerirajuće krakove. Ti rezultati ukazuju na pomlađivanje na razini tkiva. U ovom radu nije bilo moguće pokazati da se pomlađivanje odvija i na razini stanice. Ipak, mogućnost da neke populacije mogu preživjeti isključivo putem nespolnog razmnožavanja upućuje na sposobnost staničnog pomlađivanja, te bi ta mogućnost trebala biti predmet daljnjih istraživanja.
