Abstract Duboko smrzavanje (DS) sperme omogućuje korištenje vrhunskih rasplodnih
pastuha na većem broju kobila, kontrolira prenošenje spolno prenosivih bolesti,
omogućuje dobivanje potomstva od jedinki koje žive u različitim krajevima svijeta te
omogućuje pohranu genetskog materijala kroz duži vremenski period. Kvalitetan ejakulat
dobiva se od zdravog pastuha normalno razvijenih spolnih organa i s dobro izraženim
spolnim refleksima. Kontrola reproduktivnog zdravlja rasplodnih pastuha temelji se na
procjeni općeg zdravstvenog stanja životinje, a usredotočena je na spolni sustav,
lokomotorni sustav, preventivne biosigurnosne mjere, otkrivanje spolno prenosivih
zaraznih bolesti i potencijalno nasljednih genetskih poremećaja. Svaki ejakulat pastuha
podvrgava se sanitarnoj, makroskopskoj i mikroskopskoj ocjeni kako bi ustanovili
njegovu kakvoću i prikladnost za DS. Sperma potencijalnog pastuha donora mora proći
probni test DS i odmrzavanja kako bi utvrdili stopu preživljavanja spermija s obzirom na
velike varijacije u kriotoleranciji spermija između pastuha. Smrzavanje sjemena
obuhvaća uklanjanje sjemene plazme, razrjeđivanje spermija razrjeđivačem, punjenje u
pajete, smrzavanje i pohranu u tekućem dušiku. Razrjeđivači osiguravaju spermijima
hranjive tvari, stabilan pH te štite od temperaturnog i osmotskog šoka. Najčešće korišteni
krioprotektor u razrjeđivačima je glicerol koji sprječava stvaranje ledenih kristala unutar
spermija. Spermiji pastuha osobito su osjetljivi na temperaturni šok, te postoje velike
varijacije između pojedinih pastuha. Stoga se konstantno istražuju novi protokoli i
razrjeđivači koji će omogućiti optimalno preživljavanje spermija nakon DS i
zadovoljavajuće stope koncepcije. UO s DS sjemenom posebice je zastupljeno kod
sportskih konja čiji se natjecateljski kalendar često preklapa s uzgojnom sezonom.
Upravo zbog mogućnosti pohrane DS sjemena, omogućeno je polučivanje ejakulata od
rasplodnjaka i UO kobila u najpovoljnijem trenutku u odnosu na sezonu natjecanja i
početak rasplodne sezone kobila.
Abstract (english) Cryopreservation of equine semen provides the use of superior breeding stallions
on larger population of mares, controls transfer of sexually transmitted diseases, enables
offspring from horses that live in different parts of the world and storage of genetic
material for a long period of time. Good quality ejaculate is provided only from healthy
stallion with normally developed genitalia and sexual behavior. Breeding soundness
examination of donor stallions consists of general physical examination with the focus
on the reproductive organs, locomotor system, preventive biosecurity measures, and
testing for contagious venereal diseases and potentially hereditary genetic disorders. Each
ejaculate undergoes sanitary, macroscopic and microscopic evaluation, with the aim to
enhance fertility of frozen semen. Potential semen donor must undergo a freezing /
thawing quality test to determine post-thaw sperm survival rates, due to the large
variations in sperm cryotolerance between stallions. Semen freezing includes seminal
plasma removal, dilution with extenders, straw-filling, freezing and storage in liquid
nitrogen. Extenders provide sperm with nutrients, buffer and protect spermatozoa against
cold and osmotic shock. The most commonly used cryoprotectant is glycerol, which
prevents the ice formation inside spermatozoa. Stallion spermatozoa are particularly
sensitive to cold shock, and there are large variations between individual stallions.
Scientific research is constantly performed to improve protocols and extenders for
cryopreservation of equine semen with aim to improve post-thaw sperm survival with
acceptable pregnancy rate. Artificial insemination with frozen semen is especially
represented in sport horses because of the constant overlap of the competition calendar
and breeding season. Semen cryopreservation give us possibility to collect and freeze the
stallion semen and inseminate the mares in most favorable timing, regardless to the
breeding season and competition calendar.
