Abstract
Newcastleska bolest je virusna, izrazito kontagiozna zarazna bolest domaće peradi i divljih ptica koja uzrokuje visoki pobol i pomor te velike gospodarske štete zbog uginuća, mjera kontrole, suzbijanja i iskorijenjivanja. Iako u RH nije dokazana od 1994. godine, zbog velikih gospodarskih šteta koje bi mogla izazvati potrebna je brza, jednostavna i pouzdana dijagnostika newcastleske bolesti. Cilj ovog rada je istražiti mogućnost primjene metode umnažanja nukleinskih kiselina nazvane petljom posredovano izotermalno umnažanje (engl., Loop-Mediated Isothermal Amplification, LAMP) u potvrdi i apsolutnoj kvantifikaciji virusa newcastleske bolesti u kliničkim uzorcima. Reverznu transkripciju virusne RNK te amplifikaciju i detekciju ciljnih slijedova nukleinskih kiselina proveli smo u jednom koraku na stalnoj temperaturi prateći tijek reakcije pomoću RealTime PCR uređaja. Praćenje odvijanja LAMP reakcije u stvarnom vremenu omogućilo nam je približnu apsolutnu kvantifikaciju početne količine virusne nukleinske kiseline u uzorku na temelju standardne krivulje. RT-LAMP proveden u ovom radu pokazao se kao jednostavna, brza, specifična i osjetljiva metoda umnažanja kao i kvantifikacije nukleinskih kiselina.
Abstract (english)
Newcastle disease is a highly contagious viral infectious disease of domestic poultry and wild birds causing high morbidity, mortality and great economic losses due to high death rate, expensive control, suppression and eradication measures. Although the disease was not proven in Croatia since 1994., due to its devastiting effects on poultry industry there is a need for rapid, simple and realible diagnostic method. The aim of this work is to explore the posibility of using loop-mediated isothermal amplification (LAMP) in detection and absolute quantification of Newcastle disease virus in clinical samples. Reverse transcription of viral RNA, amplification and detection of target sequences was performed in one step at a constant temperature while monitoring the reaction in a RealTime PCR device. Real time monitoring of LAMP has enabled the aproximate absolute quantification of initial viral nucleic acid concentration in a sample based on the standard curve. RT-LAMP has proved itself to be a simple, rapid, specific and sensitive method of nucleic acid amplification as well as quantification.
