Abstract
Istraživanje je provedeno kako bi se utvrdila povezanost polimorfizama gena FASN i sastava masnih kiselina u mlijeku kod Holštajn-frizijske pasmine goveda. Iz ispitivanog uzorka mlijeka izdvojena je mliječna mast te je pomoću plinske kromatografije s masenom detekcijom određeni masnokiselinski sastav pomoću eksternog standard i koncentracija pojedinih masnih kiselina pomoću specifične fragmentacije. Iz mlijeka je nakon toga izolirana DNK i utvrđeni su pojedini genotipovi FASN gena za dva SNP 5848bpA/G i 5863bpT/C pomoću RFLP-PCR metode. Polimorfizam gena FASN očitovao se s dva alela (A i G) i dva genotipa AG/TC (TW/AR) i GG/CC (AR/AR). Diplotip AR/AR (GG/CC) je najučestaliji i u mlijeku se očituje većim udjelom MUFA, C18:1n9trans, C18:1n9cis i CLA. Rezultati istraživanja upućuju na važnost selekcije unutar Holštajn-frizijske pasmine krava obzirom na genotip FASN gena. Posebnu pažnju treba obratiti na genotip GG/CC (AR/AR) koji pokazuje značajan utjecaj na koncentraciju MUFA, C18:1n9trans, C18:1n9cis i CLA u mlijeku.
Abstract (english)
The study was conducted to determine the association of FASN gene polymorphisms and the milk-fat composition of Holstein-Friesian cows. Milk fat was separated and fatty acid composition was determined using gas chromatography with mass detection. Fatty acids were identified using external standard while conjugated linoleic acid was determined using characteristic fragmentation pattern. DNA was isolated from the milk samples and FASN gene genotypes were determined for two SNPs 5848bpA/G and 5863bpT/C using RFLP-PCR method. FASN gene polymorphism manifested with two alleles (A and D) and two genotype AG/TC (TW/AR) and GG/CC (AR/AR). Diplotype AR/AR (GG/CC) was the most common and the milk from the cows with that genotype had the higher concentration of MUFA, C18: 1n9trans, C18: 1n9cis and CLA. The obtained results indicate the importance of selection within the Holstein-Friesian cows based on FASN gene genotypes. Particular attention should be paid to genotype AR/AR (GG/CC) which shows significantly higher concentrations of MUFA, C18: 1n9trans, C18: 1n9cis and CLA.
