Abstract
Ovaj diplomski rad donosi pregled dosadašnjih istraživanja o dostupnim laboratorijskim metodama dijagnostike Afričke svinjske kuge. Afrička svinjska kuga zarazna je bolest koja se očituje u obliku hemoragijske septikemije i uzrokuje i do stopostotnu smrtnost svinja. Uzročnik je Asfivirus, DNK virus s dvostrukom uzvojnicom. Laboratorijske metode mogu se podijeliti na metode virusne izolacije i nalaza virusnog genoma te na serološke metode. Odabir dijagnostičkog testa ovisit će o epidemiološkoj situaciji i dostupnim resursima na tom području. U perakutnim i akutnim slučajevima odabir testa mora biti usmjeren prema izolaciji virusa na primarnim kulturama leukocitnih stanica ili kulturama stanica koštane srži. Brži i jednostavniji način dijagnosticiranja postiže se detekcijom virusnog genoma LRP reakcijom ili nalazom antigena neizravnom imunofluorescencijom. Potonje dvije metode imaju visoku osjetljivost i specifičnost. Budući da cjepivo nije dostupno, dobar pokazatelj preboljenja bolesti predstavljaju protutijela. Svinje koje prebole bolest razvijaju protutijela nakon 1. tjedna infekcije i ona perzistiraju dugo vremena. U regijama/ zemljama u kojima je bolest poprimila endemični karakter ili u onima u kojima je zabilježen proboj zaraze uzrokovan sojevima virusa niske virulencije, epidemiološka istraživanja trebaju se temeljiti na serološkim testovima. Dostupne metode za nalaz protutijela su ELISA, indirektna imunofluorescencija, indirektni imunoperoksidazni test i imunobloting test.
Abstract (english)
This thesis provides an overview of previous research on available laboratory methods for the diagnosis of African swine fever. African swine fever is a contagious disease that manifests itself in the form of hemorrhagic septicemia and causes up to one hundred percent mortality in pigs. The causative agent is Asfivirus, a double-stranded DNA virus. Laboratory methods can be divided into viral isolation and viral genome findings and serological methods. The choice of diagnostic test will depend on the epidemiological situation and the resources available in the area. In peracute and acute cases, test selection must be directed toward virus isolation on primary leukocyte cell cultures or bone marrow cell cultures. A faster and simpler way of diagnosis is achieved by detecting the viral genome by PCR reaction or finding an antigen by indirect immunofluorescence. The latter two methods have high sensitivity and specificity. Since the vaccine is not available, antibodies are a good indicator of overcoming the disease. Pigs that survive the disease develop antibodies after the 1st week of infection and they persist for a long time. In regions / countries where the disease has become endemic or in those where a breakthrough of infection is caused by low virulence virus strains, epidemiological investigations should be based on serological tests. Available methods for antibody detection are ELISA, indirect immunofluorescence, indirect immunoperoxidase assay, and immunoblotting assay.
