Abstract
Salmoneloze peradi jedna su od najčešćih bolesti na svijetu, koje se prenose hranom. No, one uzrokovane serovarom Gallinarum mogu prouzročiti velike ekonomske štete i gubitke isključivo u peradi. Zbog novih smjernica u proizvodnji peradi k slobodnom držanju na otvorenom, učestalost zaraze serovarom Gallinarum se povećava diljem svijeta. Može biti česta u jatima konzumnih nesilica koja mogu predstavljati rezervoar ovog patogena. Stoga je bitno provoditi redovitu kontrola te brzo i jednostavno moći dokazati i tipizirati patogen. U identifikaciji uzročnika, danas se najviše koristi molekularna metoda lančane reakcije polimerazom (PCR). U ovom radu proveli smo istraživanje na temelju flhB gena kako bi mogli dokazati patogen i pratiti njegovu heterogenost koristeći kombinaciju PCR metode i analize sekvenci njezinog produkta. Rezultati ukazuju na malu varijabilnost flhB gena među sojevima i nemogućnost praćenja heterogenosti sojeva na farmama temeljem sekvenci ovog gena. Zbog konzerviranosti flhB gena moguće ga je uspješno koristiti isključivo za razlikovanje od ostalih serovara salmonela.
Abstract (english)
Salmonellosis is one of the most common food-borne diseases worldwide. However, infections caused by serotype Gallinarum cause problems only in poultry, and can lead to great economic losses. Due to novel guidelines for poultry husbandry that lead to free-range production, the infection with S. Gallinarum is becoming more frequent. It is often present on laying hen farms, which act as reservoirs of this pathogen. For that reason, it is very important to do regular monitoring and be able to quickly detect and identify the pathogen. Nowadays, polymerase chain reaction (PCR) is most frequently used for identification of the causative agent. In this study, we identified and compared S. Gallinarum strains based on the flhB gene, in order to investigate the variability of the strains. The research was done using the PCR method and phylogenetic analysis of the obtained flhB sequences. The results show that flhB gene is not variable enough to detect the heterogenicity of the studied strains on farms. Since it is highly conserved, it can be used only to differentiate S. Gallinarum from other serotypes.
