Abstract
Analiza izražaja gena napravljena je tijekom in vitro diferencijacije živčanih matičnih stanica 0., 1., 3., 5. i 7. dana diferencijacije. U ovom istraživanju napravljena je analiza izražaja gena karakterističnih za živčane matične stanice (Nestin), neurone (MAP2), astrocite (GFAP) te za dva sinaptička biljega (Neuroligin1 i Synapsin1) pomoću RT-PCR-a. Rezultati RT-PCRa uspoređeni su sa rezultatima imunocitokemije. Razina izražaja Nestina najveća je tijekom nultog dana diferencijacije i nakon toga smanjuje se tijekom diferencijacije stanica. Međutim, dio stanica ostaje u nediferenciranom obliku, i čini 2,5% početne razine Nestina. Stanice, MAP2 izražavaju već nultog dana diferencijacije na razini 16,6% od maksimalnog izražaja koji se javlja sedmog dana diferencijacije. Izražaj GFAP-a javlja se trećeg dana diferencijacije i postupno raste do sedmog dana. Imnocitokemijski GFAP pozitivne stanice vidljive su tek petog dana u 1% prebrojanih stanica. Oba sinaptička biljega pozitivna su već nultog dana diferencijacije i razina njihovog izražaja postupno se povećava za 70% od početne vrijednosti, pri čemu je razina Neuroligina1 jača u odnosu na Synapsin1.
Abstract (english)
Analysis of genes expression during in vitro differentiation of neural stem cells 0, 1, 3, 5 and 7 days of differentiation were made. In this study expression of genes typical for neural stem cells (Nestin), neurons (MAP2), astrocytes (GFAP) and two synaptic markers (Neuroligin1 and Synapsin1) were analysed using RT-PCR. Results of RT-PCR were compared with the results of immunocytochemistry. The highest Nestin expression was detected on the day zero and expression decreased with cell differentiation. Some cells remained undifferentiated and accounted 2.5% of the initial Nestin expression. MAP2 expression on the day zero of differentiation was 16.6% of the maximum expression occurring on the seventh day of differentiation. GFAP expression was observed on the third day of differentiation and gradually increased. On the fifth day of differentiation immunocytochemical analysis showed that 1% of the counted cells were GFAP positive. Both synaptic markers were positive already at day zero of differentiation and their expression gradually increased to 70% of the initial value; Neuroligin1 expression was stronger than Synapsin1 expression.
