Abstract (english)
Citomegaloviruses (CMVs) are DNA viruses that can cause severe disease in immunocompromised persons. Very early during infection, CMVs extensively reorganize membranous compartments of the host cells which finally results with formation of virion assembly compartment (VAC). Since Arf GTPases are one of the major regulators of cellular membrane dynamics we hypothesized that CMVs manipulate Arf protein system in order to ensure favorable conditions for their replication. To address this hypothesis, we infected Balb 3T3 cells with murine CMV (MCMV), since organization structure of human and murine VAC is similar but at the same time murine MCMV replication cycle is much shorter and therefore more suitable for studying. In MCMV infected cells expression of class I (Arf1, Arf3), clas II (Arf4, Arf5) and clas II (Arf6) proteins was determined with Western blot and immunofluorescent microscopy in the early phase of the infection. To determine their intracellular localization, each of the Arf proteins was colocalized with markers of early and late endosomal compartments, markers of Golgi complex, as well as with different early phase viral proteins. Our results indicate that Arf proteins are overexpressed (Arf1-5) or overactivated (Arf6) during the early phase of CMV infection. They also change their intracellular localization and are accumulated in the inner part of VAC where they colocalize with markers of early and recycling endosomes. Better understanding of VAC formation and the role that Arf proteins play in that process may enable development of new antiviral and therapeutic strategies.
