Abstract
Glukozinolati su jedinstvena skupina tioglukozida s identičnom osnovnom strukturom, karakteristični za porodicu Brassicaceae, kojoj pripada i brokula. Razlika u sadržaju glukozinolata u povrću ovisi o genotipu, klimatskim uvjetima, godišnjem dobu uzgoja i agronomskim uvjetima. Pomoću HPLC smo analizirali ukupne i pojedinačne glukozinolate u dvije sorte brokule Parthenon i Ironman. Glukozinolati su kvantificirani Standardnom metodom za određivanje glukozinolata koja uključuje ekstrakciju s metanolom, enzimsku desulfataciju i reverzno-faznu tekućinsku kromatografiju (RP-LC). Identifikacija eluiranih desulfoglukozinolata je napravljena prema redoslijedu eluiranja s kromatografske kolone i analize UV-spektara. Ukupni sadržaj glukozinolata u ispitivanim sortama bio je sličan (9.96 i 9.37 μmol g-1 s.t. za Parthenon, odnosno Ironman). U obje sorte prevladavaju indolni glukozinolati. U sorti Ironman je zamijećen veći udio glukorafanina, prekursora sulforafana, nego kod sotre Parthenon.
Abstract (english)
Glucosinolates are a uniform group of thioglucosides with an identical core structure that are characteristic for Brassicaceae family, which include broccoli. The difference in the content of glucosinolates in vegetables depends on the genotype, climate conditions, season of breeding, and agronomic conditions. We analyzed total and individual glucosinolates in two broccoli cultivars Parthenon and Ironman by HPLC. Glucosinolates were quantified by Standard method for glucosinolates determination and it includes extraction with methanol, enzymatic desulphation and Reversed-Phase Liquid Chromatography (RP-LC). The identification of desulphoglucosinolates eluted was made according to the elution order on column chromatography and also by UV-spectar analyses. The total glucosinolate content in investigated cultivars were similar (9.96 and 9.37 μmol g-1 d.w. for Parthenon and Ironman, respectively). The most predominate compound in both cultivars were indole glucosinolates. In Ironman cultivar the higher content of glucoraphane, a precursor of sulforaphane, then in Parthenon was observed.
