Abstract
Zbog važnosti kvasca Saccharomyces cerevisiae u biotehnološkoj industriji, izvršena su mnoga ispitivanja utjecaja pojedinih elemenata na njegov rast, a jedan od važnijih ispitivanih elemenata je bakar. Bakar je metal s dvojakom funkcijom u metabolizmu kvasca S. cerevisiae. On je esencijalan za kvasac jer je građevni dio jedinica respiratornog lanca, ali pokazuje toksičnost pri višim koncentracijama. U ovom radu istraživan je toksični utjecaja bakra na rast sojeva kvasca S. cerevisiae različitog ploiditeta te pri različitim uvjetima, tj. pri različitoj pH vrijednosti i sastavu podloge. Prema dobivenim rezultatima, možemo ustvrditi da je toksičnost bakra veća na kemijski definiranim podlogama nego na kompleksnim. Također, toksičnost bakra je veća pri neutralnom pH nego pri nižim pH vrijednostima. Razlika u toksičnosti bakra prema diploidnom i haploidnom soju kvasca nije uočena, ali je zabilježen bolji rast soja disomika za osmi kromosom od haploidnog i diploidnog soja na podlogama bez izvora sulfata.
Abstract (english)
Because the yeast Saccharomyces cerevisiae is one of the most important industrial microorganisms, a lot of research was conducted on the impact of various elements on its growth. Among these, one of the most important elements is copper which has a dual biological role in the metabolism of the yeast S. cerevisiae. Copper is essential for yeast cells because it is a component of respiratory chain units, but it becomes toxic at higher concentrations. In this work the toxic effect of copper on the growth of yeast strains of different ploidities was studied. This effect was also investigated under several conditions, i.e. the composition and the pH of the medium. As the results show, the copper exhibits higher toxicity when the cells are grown on synthetic complete medium instead of the complex medium. Also, the toxicity of copper is higher at neutral pH than at lower pH values. There was no difference in toxic effect of copper on diploid and haploid yeast strains. However, yeast strain which was disomic for the chromosome VIII tolerated higher concentrations of copper than the both haploid and diploid strain if the medium without the addition of sulphate salts was used.
