Abstract
Cilj ovog rada bio je ispitati in vitro preživljavanje probiotičke bakterije Bifidobacterium animalis subsp. lactis BB-12 u simuliranim uvjetima gastrointestinalnog trakta, istražiti autoagregacijska i koagregacijska svojstva s drugim sojevima bakterija mliječne kiseline Lactobacillus helveticus M92 i Enterococcus faecium L3 te potencijalno patogenim mikroorganizmima Escherichia coli 3014 i Salmonella enterica serovar Typhimurium FP1 i usporediti rezultate s rezultatima dobivenim s drugom probiotičkom bakterijom Bifidobacterium longum BB536. Obje probiotičke bakterije pokazuju visoku otpornost prema djelovanju pepsina te dobro podnose visoke koncentracije vodikovih iona, dok je soj B. animalis subsp. lactis BB-12 pokazao bolju toleranciju na djelovanje enzima gušterače i žučnih soli. Bakterijski soj B. animalis subsp. lactis BB-12 pokazuje veći postotak autoagregacije kao i bolja koagregacijska svojstva s drugim sojevima bakterija mliječne kiseline od B. longum BB536. Kod bakterijskog soja B. animalis subsp. lactis BB-12 je ustanovljena relativno visoka koagregacija s potencijalno patogenim test-mikroorganizmima, dok su kod soja B. longum BB536 zabilježeni znatno niži postoci koagregacije. Prema dobivenim rezultatima, bakterija B. animalis subsp. lactis BB-12 zadovoljava postavljene kriterije i može obavljati probiotičku funkciju u tijelu domaćina
Abstract (english)
The aim of this work was to characterize in vitro survival of probiotic bacteria Bifidobacterium animalis subsp. lactis BB-12 in simulated conditions of gastrointestinal tract, and explore ability of auto-aggregation and co-aggregation with other lactic acid bacteria Lactobacillus helveticus M92 and Enterococcus faecium L3 and potentially pathogenic microorganisms Escherichia coli 3014 and Salmonella enterica serovar Typhimurium FP1, and compare the results with results obtained with other probiotic bacteria Bifidobacterium longum BB536. Both probiotic bacteria show high resistance to the activity of pepsin and to high concentration of hydrogen ions, but strain B. animalis subsp. lactis BB-12 has shown better resistance to the activity of pancreatic enzymes and to bile salts. Bacterial strain B. animalis subsp. lactis BB-12 show higher percentage of auto-aggregation and the ability of co-aggregation with other lactic acid bacteria than B. longum BB536. At bacterial strain B. animalis subsp. lactis BB-12 was detected a relatively high co-aggregation with potentially pathogenic microorganisms, while B. longum BB536 showed significantly lower percentages of co-aggregation. Binding tests showed that B. animalis subsp. lactis BB-12 satisfies set requirements and can perform probiotic function in the body of the host.
