Abstract
Niskotemperaturna eutektička otapala (NADES) sve se češće koriste za zelenu ekstrakciju biološki aktivnih spojeva. Njihova fizikalno-kemijska svojstva mogu se dizajnirati za posebne namjene tako da zadovoljavaju tehnološke i ekonomske zahtjeve industrije. Cilj ovog rada bio je dizajnirati i odabrati NADES pomoću programa COSMOtherm za ekstrakciju polifenola. Polifenoli su ekstrahirani iz komine grožđa, nusproizvoda vinske industrije, bogate biološki aktivnim spojevima. Odabrano je otapalo betain:saharoza te je određena ukupna koncentracija polifenola Folin-Ciocalteu reagensom i uspoređena sa ekstraktima u konvencionalnim otapalima (metanolu i etanolu). Pojedini polifenoli identificirani su HPLC metodom. Ispitana je mogućnost ekstrakata za inhibiciju aktivnosti kolagenaze te je analiziran utjecaj ekstrakata na staničnu liniju ljudskih keratinocita (engl. Cultured Human Keratinocyte, HaCaT). Na temelju dobivenih rezultata može se zaključiti da NADES otapala mogu zamijeniti konvencionalna otapala, a zbog uočene proliferacije HaCaT stanica ekstrakti imaju mogućnost primjene u kozmetičkoj industriji.
Abstract (english)
Natural deep eutectic solvents (NADES) are increasingly being used for the green extraction of biologically active compounds. Their physicochemical properties can be designed for special purposes so that they fulfill the technological and economic requirements of the industry. The aim of this work was to design and select NADES using the COSMOtherm program for the extraction of polyphenols. Polyphenols are extracted from grape pomace, a byproduct of the wine industry, rich in biologically active compounds. The solvent betaine:sucrose was chosen and the total concentration of polyphenols was determined with the Folin-Ciocalteu reagent and compared with extracts in conventional solvents (methanol and ethanol). Individual polyphenols were identified by the HPLC method. The ability of the extracts to inhibit collagenase activity was tested, and the influence of the extracts on the human keratinocytes cell line (HaCaT) was analyzed. Based on the obtained results, it can be concluded that NADES solvents can replace conventional solvents, and due to the observed proliferation of HaCaT cells, the extracts have the possibility of application in the cosmetic industry.
