Abstract
Proteini su makromolekule koje se nalaze u svim živim organizmima i sudjeluju u svim fiziološkim procesima, što ih čini predmetom istraživanja u različitim područjima istraživanja poput biologije, biokemije, biotehnologije, medicine, kao i u farmaceutskoj industriji. Navedena istraživanja mogu biti usmjerena ka karakterizaciji nepoznatih proteina (ponajprije njihovu identifikaciju) ili kvantifikaciju. Identifikacija proteina omogućuje daljnje određivanje svojstava proteina kao što su potvrda strukture (primarne, sekundarne, tercijarne i/ili kvaterne) posttranslacijskih modifikacija, fizikalno-kemijskih svojstava (pI, molekulska masa), ali i određivanje njihove biološke funkcije i lokalizacije. Određivanje molekulske mase posebno je važno kod karakterizacije pročišćenih proteina od interesa (npr. enzima i biofarmaceutika). Rad će se usmjeriti na određivanje molekulske mase odabranim fizikalno-kemijskim metodama. Iako postoje brojne metode, fokus će biti na analitičkom centrifugiranju, elektroforezi u gelu te spektrometriji masa kao najsuvremenijoj i najsofisticiranijoj metodi.
Abstract (english)
Proteins are macromolecules essential for life participating in numerous physiological processes, which makes them the subject of study in various research fields such as biology, biochemistry, biotechnology, medicine and the pharmaceutical industry. The research can be focused on the characterization of unknown proteins (preferably their identification) and/or quantification. Protein identification enables further determination of protein properties such as structure determination (e.g. amino acid sequence, secondary, tertiary and quaternary structure, post-translational modification), physicochemical properties (pI, molecular weight) or biological function. Molecular weight determination is crucial for purified proteins of interest (e.g. enzymes or biopharmaceuticals). Although there are numerous methods, this work will be focus on the determination of molecular weight by selected physicochemical methods: analytical centrifugation, gel electrophoresis, and finally mass spectrometry as the most modern and sophisticated method.
