Abstract | U ovom radu istraživan je utjecaj dodatka različitih induktora (bakrov sulfat, ferulična kiselina, siringična kiselina, vanilinska kiselina, veratrilni alkohol) na aktivnost lakaze i mangan-peroksidaze tijekom sedam dana uzgoja T. versicolor na kukuruznoj silaži u malom mjerilu s početnom masom supstrata od 50 g. Najveće aktivnosti oba enzima postignute su 7. dan provedbe pokusa u pokusima kada je kao induktor korištena 0,1 mol dm\(^{-3}\) otopina CuSO\(_{4}\), pri čemu je volumna aktivnost lakaze iznosila 1389,1 U dm\(^{-3}\), a mangan-peroksidaze 209,1 U dm\(^{-3}\) Napravljeno je uvećanje procesa u bioreaktoru s pliticama, dimenzija 75 x 154 x 70 cm, sa šest plitica dimenzija 50 x 5 x 40 cm. Proces je proveden pri sljedećim početnim uvjetima: m\(_{kukuruzne silaže}\) = 600 g (po svakoj plitici), vlažnost supstrata = 82,30 ± 0,26 %, visina sloja supstrata = 3 cm, veličina čestica supstrata = 1-2 cm, masa inokuluma: 200 g, protok zraka = 2,5 dm\(^{3}\) min\(^{-1}\), T = 27 °C, t = 5 - 7 dana, c CuSO\(_{4}\) = 0,1 mol dm\(^{-3}\). Najveće aktivnosti oba enzima postignute su 7. dan provedbe pokusa, pri čemu je volumna aktivnost lakaze iznosila 722,1 U dm\(^{-3}\), a mangan-peroksidaze 140,1 U dm\(^{-3}\). |
Abstract (english) | In this work, the influence of various inducers (copper sulphate, ferulic acid, syringic acid, vanillic acid, veratryl alcohol) on activity of laccase and manganese-peroxidase, during 7 days solid-state small-scale cultivation of T. versicolor on corn silage with the initial substrate mass of 50 g, was investigated. The highest enzymes activities were achieved on the seventh day using 0.1 mol dm\(^{-3}\) of CuSO\(_{4}\) as an inducer, and were 1389,1 U dm\(^{-3}\) and 209,1 U dm\(^{-3}\) for laccase and manganese-peroxidase, respectively. The scale-up of the process was done in the tray bioreactor (75 x 154 x 70 cm) comprised of six trays (50 x 5 x 40 cm). The process was carried out at the following initial process conditions: m\(_{corn silage}\) = 600 g (per tray), substrate humidity of 82,30 ± 0,26%, substrate layer height = 3 cm, substrate particle size = 1- 2 cm, inoculum mass = 200 g, airflow = 2.5 dm\(^{3}\) min\(^{-1}\), T = 27 °C, t = 5 - 7 days, cCuSO\(_{4}\) = 0,1 mol dm\(^{-3}\). The highest enzymes activities were achieved on the seventh day and were 722.1 U dm\(^{-3}\) and 140.1 U dm\(^{-3}\), for laccase and manganese-peroxidase, respectively. |
