Abstract
Cilj ovog završnog rada bio je provesti elektroforetsku analizu proteina (SDS-PAGE) membrana jaja pripravljenih različitim postupcima obrade ljuske jaja kao otpada poljoprivredno-prehrambene industrije. U prvom koraku su membrane jaja ispitane na optimalno vrijeme termičke ekstrakcije pomoću pufera za obradu proteina za SDS-PAGE tijekom 30, 60 i 120 minuta, pri čemu je ustanovljeno da je za potpunu ekstrakciju proteina membrana jaja dostatno vrijeme od 30 minuta termičke obrade. U sljedećem koraku su membrane jaja ispitane na razliku u sastavu/intenzitetu proteinskih vrpci ekstrahiranih iz membrana jaja tijekom 30 minuta. SDS-PAGE analiza je pokazala da se najviše proteinskih vrpci membrana jaja detektira u membranama jaja pripravljenim kiselinskom obradom ljuske jaja sa 10 % octenom i 15 % o-fosfornom kiselinom.
Abstract (english)
The aim of this undergraduate thesis was to perform SDS-PAGE analysis of eggshell membrane (ESM) proteins of ESM’s prepared by different eggshell waste processing procedures. In the first step, eggshell membranes were analysed for the optimal time of thermal extraction of ESM proteins using SDS-PAGE of samples treated by SDS-PAGE treatment buffer for 30, 60 and 120 minutes, where it was found that 30 minutes of thermal treatment is sufficient for complete protein extraction of the eggshell membrane proteins. In the next step, the eggshell membranes were analysed for differences in the composition/intensity of the protein bands extracted from the eggshell membranes during 30-minute treatment with SDS-PAGE treatment buffer. SDS-PAGE analysis showed that the most protein bands of eggshell membranes were detected in eggshell membranes prepared by acid treatment of eggshell waste with 10% acetic and 15% o-phosphoric acid.
